乌梅丸对大鼠胃癌及癌前病变中端粒酶和PCNA表达的影响

目的：探讨乌梅丸对胃癌及癌前病变中端粒酶和PCNA的表达的影响。方法：建立大鼠胃黏膜癌病变模型,采用TRAP-ELISA方法检测大鼠胃黏膜端粒酶活性,检测增殖细胞核抗原（PCNA）的表达。结果：低剂量乌梅丸组（简称低剂量组）中重度异型增生病变数少于模型组,存在统计学差异（P〈0.05）,高剂量乌梅丸组（简称高剂量组）中重度异型增生病变数少于低剂量组,存在显著性差异（P〈0.01）;低剂量组端粒酶活性低于模型组,存在统计学差异（P〈0.05）,高剂量组端粒酶活性低于低剂量组,存在统计学差异（P〈0.05）;低剂量组PCNA阳性表达率低于模型组,存在统计学差异（P〈0.05）,高剂量组PCNA阳性表达率低于低剂量组,存在统计学差异（P〈0.05）。结论：乌梅丸能抑制端粒酶活性和PCNA的表达,并且呈剂量依赖性,这可能是乌梅丸干预胃癌及癌前病变的机制之一。

The Impact of Wumeiwan on Telomerase Activity and Expression of PCNA in Gastric Cancer and Precancerous Lesions of Rats

Objective： To explore the impact of Wumeiwan on telomerase activity and expression of PCNA in rats＇ gastric cancer and precancerous lesions.Methods： establish rat model of gastric cancer lesions,use the TRAP-ELISA method to detect the telomerase activity in rat gastric mucosa,detect proliferating cell nuclear antigen（PCNA） expression.Results： The low-dose Wumeiwan group（low-dose group） with severe dysplasia in lesions is less than model group,there is a difference in statistics（P 0.05）,high-dose Wumeiwan group（high-dose group） with severe dysplasia in lesions is less than low-dose group,there is a significant difference（P0.01）,telomerase activity in low-dose group lower than model group,there is a significant difference（P0.05）,telomerase activity of high dose group lower than the low-dose group,there is a difference in statistics（P0.05）,the positive expression rate of PCNA in low-dose group is lower than model group,there is a difference in statistics（P0.05）,the positive expression rate of PCNA in high-dose group was lower than the low-dose group,there is statistical differences（P0.05）.Conclusion： Wumeiwan can inhibit telomerase activity and PCNA expression,there is a dose-dependent manner,it may be one of the mechanisms that Wumeiwan interfere with gastric cancer and precancerous lesions.
