中肺合剂对Lewis肺癌小鼠肿瘤组织血管生成的影响

摘    要:目的:观察中肺合剂对Lewis肺癌小鼠肿瘤组织血管生成的影响。方法:取C57BL/6小鼠50只,接种Lewis肺癌瘤株后,随机分阴性对照组(0.9%生理盐水),中肺合剂低、中、高剂量组(12.7、25.4、50.8g.kg-1),阳性对照组(CTX组,30mg.kg-1),每组10只。观察12天停药。无菌条件下取肿瘤组织,采用EnVision法检测肿瘤组织血管内皮生长因子(VEGF)和CD34蛋白的表达情况,并计数微血管密度(MVD)。结果:中肺合剂能降低肿瘤组织中血管内皮生长因子(VEGF)的表达和微血管密度(MVD),VEGF表达和MVD之间呈正相关(P〈0.01)。其中以中、高给药剂量组抑制作用显著,与生理盐水组相比有显著差异(P〈0.01)。低剂量组与生理盐水组相比无显著差异(P〉0.05)。结论:中肺合剂能抑制Lewis肺癌小鼠肿瘤组织新生血管的形成,降低Lewis肺癌小鼠肿瘤组织中血管内皮生长因子(VEGF)的表达和微血管密度(MVD),以中、高剂量组最明显。

Abstract:Objective: To observe the effects of Zhongfei Mixture(ZM) on angiogenesis in tumor tissues from the mice bearing Lewis lung carcinoma.Methods: Fifty C57BL/6 mice bearing Lewis lung carcinoma were divided into 5 groups:the saline control group,the low,mid and high-dose groups of ZM and Cyclophosphamide(CTX) group.Tumor tissues were isolated after 12 days followed by the expression detection of vascular endothelial growth factor(VEGF),CD34 protein and the microvessel density(MVD) in tumor tissues by EnVision two-step immuno-histochemistry staining.Results: ZM could down-regulate the expression level of VEGF and CD34 in tumor tissues from the mice bearing Lewis lung carcinoma.There was a significantly positive correlation between VEGF and MVD.Compared with saline control group,the effects of the mid and high-dose groups on the two proteins showed significantly different in statistics(P0.01).Conclusion:ZM could depress vascular growth in tumor tissues from the mice bearing Lewis lung carcinoma.The effects of the mid and high-dose groups were superlative.
