温化蠲痹方诱导胶原诱导性关节炎大鼠滑膜细胞凋亡的研究

目的：研究中药温化蠲痹方诱导胶原诱导性关节炎（CIA）大鼠滑膜细胞凋亡作用。方法：选用健康雄性Wistar大鼠,随机分为正常对照组（n=10,简称正常组）、造模组,造模组采用牛Ⅱ型胶原乳剂于大鼠尾根部注射,建立CIA模型。造模成功后随机分为模型组（n=10）、温化蠲痹方组（n=10,简称中药组）、MTX组（n=10）。正常组、模型组、中药组按22.9g·kg-1·d-1剂量分别灌胃生理盐水、温化蠲痹方,均每日1次,MTX组按0.78mg·kg-1·w-1剂量灌胃MTX混悬液,每周1次。给药30天后,运用透射电镜和流式细胞术检测大鼠滑膜细胞凋亡。结果：与模型组比较,治疗后中药组和MTX组大鼠关节炎指数（AI）评分明显降低（P〈0.01）;与治疗前比较,中药组和MTX组大鼠AI评分均明显降低（P〈0.01）。透射电镜显示,中药组大鼠滑膜细胞凋亡较模型组大鼠滑膜细胞凋亡明显。流式细胞术检测显示,与正常组比较,模型组大鼠滑膜细胞凋亡率明显降低（P〈0.05）;与模型组大鼠比较,中药组和MTX组大鼠滑膜细胞凋亡率明显升高（P〈0.01）。结论：CIA大鼠滑膜细胞凋亡减少,诱导滑膜细胞凋亡可能是中药温化蠲痹方治疗CIA的作用机制之一。

The Study on Inducing Apoptosis of Synoviocytes in Rats with Collagen-Inducing Arthritis by Wenhua Juanbi Recipe

Objective：To study the effects on inducing apoptosis of synoviocytes in rats with collagen-Inducing arthritis（CIA） by Wenhua Juanbi recipe.Methods：Healthy Wistar rats were randomly divided into making model group and control group（n=10）.Rats of making model group were injected 0.5ml BCⅡ（type Ⅱcollegan of bovine,BCⅡ） emulsion in the tail to establish CIA model.The succeeded CIA rats were randomly divided into three groups,those were model group（n=10）,Wenhua Juanbi recipe group（n=10,Chinese heres group for short）,MTX group（n=10）.Rats of control group,model group and Chinese heres group were given Physiological saline and Wenhua Juanbi recipe respectively by stomach rearing according to 22.9g·kg-1·d-1,rats of MTX group were given MTX liquor according to 0.78mg·kg-1·w-1 in the same way.After treatment 30 days, The apoptosis of synoviocytes was determined by flow cytometric analysis and electron microscope.Results：AI score of Chinese heres group and MTX group after treatment was lower than that of model group（P0.01）.AI score of those lower than before treatment（P0.01）.The apoptosis of synoviocytes in Chinese medicine group was more obvious than in model group by electron microscope.The apoptosis of synoviocytes in Chinese medicine group was was lower significantly than in control group by Flow cytometer（P0.05）.The apoptosis of synoviocytes in Chinese medicine group and MTX group was higher significantly than in model group（P0.01）.Conclusion：The apoptosis of synoviocytes reduces in CIA rats.It’s one of possible mechenism of treating CIA that Wenhua Juanbi recipe induces the apoptosis of synoviocytes.
