生津合剂治疗大鼠急性放射性食管损伤的实验研究

摘    要:目的:探讨生津合剂治疗急性放射性食管损伤的作用机制。方法:用加速器一次性照射43Gy诱导SD大鼠发生食管炎,建立大鼠急性放射性食管损伤模型,按实验设计处理各组大鼠,研究使用生津合剂后大鼠一般状况,如体重、摄食量的影响,观察相关的血清炎性细胞因子(TNF-α,IL-1,IL-6)水平、外周血白细胞计数及分类变化,制作食管病理切片并评价其病理改变。结果:成功建立大鼠急性放射性食管炎模型,实验结果证明辐射导致放射性食管炎大鼠外周血白细胞总数、淋巴细胞计数及百分数降低,中性粒细胞百分比升高,血清炎性细胞因子(TNF-α,IL-1,IL-6)水平升高;与模型组比较,中药生津合剂对大鼠放射导致的食管损伤有一定治疗作用,能明显改善大鼠一般状况,提高大鼠外周血淋巴细胞计数及百分数(P〈0.05),降低中性粒细胞百分比(P〈0.05),下调血清炎性细胞因子(TNF-α,IL-1,IL-6)水平(P〈0.05);中药高剂量组食管黏膜组织病理积分较模型组低,有统计学差异(P〈0.05)。结论:中药生津合剂对大鼠的急性放射性食管炎具有一定治疗作用。

Abstract:Objective :To observe the mechanism of action about Shenjin mist in preventing and treating acute esophageal radioactive injury. Methods : The animal model of acute esoghageal radioactive injury in SD rats was induced by single irradiation of 43Gy gamma ray locally. Deal with all rats according to the experimental design. Investigate the changes of physiological conditions (body weight, food intake)after the rats treated with shenjin mist . And the serum levels of tumor necrosis factor - α ( TNF - α), interleukin - 6 ( IL - 6 ) and interleukin - 1 ( IL - 1 ), the leucocyte count and different in blood, the esophageal pathological changes were detected in the experiment. Results :The animal model of acute esoghageal radioactive injury was successfully established. The experimental results manifested that leukocyte count , lymphocyte differential count and absolute count of experimental rats were decreased after radiation, while neutrophil differential count in blood, eytokine such as TNF-α, IL- 6 and IL- 1 in serum increased. Compared with model group, Shenjin mist was effective in treating radiation - induced esophagitis of rats to a certain extent. Shenjin mist could improve the whole conditions, increase lymphocyte differential count and absolute count , decrease neutrophil differential count in blood , down - regulate the level of TNF - α, IL - 6 and IL - 1 ( P 〈 0.05 ). The score of esoghagus mucosa pathologic changes of high - dosage of Shenjin mist group was lower than that in model group. ( P 〈 0.05 ). Conclusion : Shenjin mist possesses preventive and therapeutic effect on the rats with radioactive injurys of esophageal to a certain exent.
