补肾强肝活血法对缺血再灌注大鼠脑损伤颅内Glu、SOD、MDA的影响

摘    要:目的:探讨补肾强肝活血法对缺血再灌注大鼠脑组织谷氨酸(glutamate,Glu)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)的影响。方法:采用线栓法复制大脑中动脉阻塞(MCAO)再灌注大鼠模型,以尼莫地平为对照,用补肾强肝活血法经验方“回春偏瘫方”治疗14天后观察其对缺血再灌注大鼠脑组织Glu、SOD和MDA含量的影响。结果:与假手术组比较,模型组大鼠脑组织出现明显Glu和MDA含量升高(P〈0.05),SOD含量降低(P〈0.05);与模型组比较,西药组和回春偏瘫方中、高剂量组脑组织Glu有不同程度的下降,SOD均有不同程度的上升(P〈0.05),但从MDA水平看,西药组和回春偏瘫方中、高剂量组有明显下降(P〈0.05),但回春偏瘫方低剂量组与模型组比较没有明显差别(P〉0.05)。结论:补肾强肝活血法可通过降低Glu和MDA水平,提高SOD含量,减轻兴奋性谷氨酸所致细胞毒性和自由基损伤作用,可有效减轻缺血性损伤。

Effects of Decoction for Rejuvenating Hemiplegia on Glu,SOD,and MDA in Brain of Experimental Ischemia-reperfusion Rats

Abstract:Objective : To investigate the effects of Decoction for Rejuvenating Hemiplegia on Glutamate ( Glu), Superoxide Dismutase (SOD) and Malondialdehyde (MDA) in the brain of experimental ischemia-reperfusion rats under guidance of the theory of tonifying liver and kidney to activate blood circulation. Method : The models of middle cerebral artery occlusion (MCAO) were made by ligating right arteria cerebri media of rats. Nimodipine was as the control. And Decoction for Rejuvenating Hemiplegia treated treatment groups. The treatment course included 14 days. Then SOD, MDA and Glu in brain were detected. Result:SOD decreased but Glu and MDA increased more in model group than in sham operation group ( P 〈 0. 05 ). SOD raised but Glu reduced more in control group, high dose group and middle dose group than in model group ( P 〈 0. 05 ). MDA reduced in control group, high dose group and middle dose group (P 〈 0. 05 ), while no significant difference showed up between low dose group and model group ( P 〉 0. 05 ). Conclusion : Decoction for Rejuvenating Hemiplegia can raise SOD and reduce Glu and MDA. So it alleviates cytotoxicity and free radical injury induced by irritable aminoglutaminic acid, so as to relieves cerebral ischemic injury.
