黄芪对大鼠胃溃疡愈合过程中表皮生长因子及其受体表达的影响

摘    要:目的:探讨黄芪治疗溃疡病的分子生物学机制。方法:将72只Wistar大鼠随机分为正常组、黄芪组、洛赛克组、模型组。除正常组外,其他各组采用Okabe乙酸涂抹法制作大鼠实验性胃溃疡模型,在造模结束后24h,各组分别以1mL/100g体重给药或给生理盐水。在第7、14、21天分批处死大鼠。观测大鼠溃疡愈合指数、光学显微镜下溃疡愈合质量、免疫组化方法测EGF、EGFR的表达情况。结果:(1)黄芪可以促进溃疡愈合。(2)黄芪组的愈合质量优于洛赛克组。(3)溃疡形成后各组EGF、EGFR表达增加,并且随着溃疡愈合其表达逐渐减弱,黄芪组的表达水平均优于其他组别,洛赛克组则与模型组差异不显著。结论:黄芪治疗胃溃疡的作用机理可能是通过促进EGF、EGFR的表达,使胃黏膜上皮组织、腺体结构较好地修复和重建,从而促进溃疡愈合并且提高溃疡愈合质量。

Abstract:Objective:To investigate the molecular biological mechanism of Chinese herbal milk veteh.Methods:Seventy-two rats were randomly divided into four groups.Gastric ulcer models were make by serosal application of acetic acid.Rats were gavaged once per day with dosage of 1 mL/100 g or normal saline.Rats were anesthetized with nembutal and stomachs were removed after 7,12 or 21 days.Gastric ulcer index(GUI),histological assessment of healing quality of ulcer,express of EGF and EGFR in gastric mucosa were determined.Results:Milk veteh could improve the healing of gastric ulcer.Express of EGF and EGFR was enhanced in ulcer models and decreased in healing phase,the express in the milk veteh group at different phases were the highest.Conclusion:The expresses of EGF and EGFR were impoved in ulcer by milk veteh.It suggested the main mechanism effect of milk veteh on ulcer.
