四逆汤配方颗粒和饮片煎剂抗心肌损伤作用的比较研究

摘    要:目的:比较四逆汤配方颗粒及其传统饮片煎剂对心肌细胞缺氧损伤模型和大鼠离体心脏缺血再灌注(I/R)损伤的保护作用。方法:建立H 2O 2诱导H9c2细胞氧化损伤模型,四逆汤配方颗粒及饮片煎剂干预后,MTT法检测细胞存活率,试剂盒法检测细胞内活性氧(ROS)、脂质过氧化物(MDA)及乳酸脱氢酶(LDH)水平。建立离体SD大鼠心脏I/R损伤模型,观察I/R期间四逆汤配方颗粒及饮片煎剂干预对左心室舒张末压(LVEDP)、心率(HR)和室内压最大上升速率(+dp/dtmax)及室内压最大下降速率(-dp/dtmax)的影响。结果:2.5、5.0 g·L-1四逆汤配方颗粒和2.5、5.0 g·L-1四逆汤饮片煎剂预处理均显著提高H 2O 2诱导氧化损伤细胞的存活率(P<0.01,P<0.05)。与H 2O 2组相比,2.5、5.0 g·L-1四逆汤配方颗粒和饮片煎剂均降低氧化损伤细胞内ROS水平(P<0.05,P<0.01)、MDA含量(P<0.01)以及LDH释放率(P<0.01),相同浓度下两者之间无统计学差异。四逆汤配方颗粒和饮片在0.5、2.0、4.0 g·L-1剂量下均可改善I/R所致的LVEDP、HR和±dp/dtmax变化,两者之间无统计学差异。结论:四逆汤配方颗粒及其饮片煎剂对心肌细胞缺氧和心脏I/R损伤均具有一定的保护作用,两者药效无明显差异。

Abstract:Objective:To investigate and compare the protective effects of Sini Decoction formula granules and its traditional medicinal slices on hypoxia injury in myocardial cell model and ischemia reperfusion(I/R)injury in rats’isolated heart.Methods:Hydrogen peroxide(H2O2)induced oxidative damage cell model was established in myocardial H9c2 cells.After cultured with Sini Decoction formula granules or prepared slices,cell survival rates were determined by MTT method,and intracellular reactive oxygen species(ROS),lipid peroxide(MDA)content and lactate dehydrogenase(LDH)release rate were detected by kits.The I/R injury models of isolated SD rats’heart were established.During the period of I/R,Left ventricular end diastolic pressure(LVEDP),heart rate(HR)and maximum rate of increase of indoor pressure(+dp/dtmax)and maximum rate of decrease of indoor pressure(-dp/dtmax)were observed after cultured with Sini Decoction formula granules and prepared slices.Results:Pretreatment with 2.5,5 g·L-1 Sini Decoction formula granules or 2.5,5 g·L-1 Sini Decoction pieces could improve the cell viability of myocardial cells induced by H2O2(P<0.01,P<0.05).Compare with the H2O2 group,2.5,5 g·L-1 Sini Decoction formula granule and prepared slices reduced the intracellular ROS level(P<0.05,P<0.01),MDA content(P<0.01)and LDH release rate(P<0.01),and there was no statistical difference between the two under the same concentration.Both the formula granules and slices of Sini Decoction at doses of 0.5,2.0,4.0 g·L-1 could improve the changes of LVEDP,HR and±dp/dtmax caused by I/R,and there was no statistical difference between the two.Conclusions:Sini Decoction formula granule and its decoction pieces have certain protective effects on myocardial cell hypoxia and cardiac I/R injury,and there is no significant difference between the two effects.
