基于中医“治未病”理论探讨骨蚀灵胶囊预防激素性股骨头坏死的实验研究

目的:探讨激素诱导股骨头坏死发生发展过程中脂质代谢的变化规律,以及早期给予中药干预的作用效果及机制,寻求有效预防激素性股骨头坏死的新思路。方法:通过激素诱导股骨头坏死实验兔模型,实验分正常组、模型组、西药组、中药组,分组给药,于给药后3、5周测定各组家兔血脂;同时观察局部股骨头骨组织形态学的相应改变。结果:3周时,与正常对照组比较,模型组、中药组、西药组兔血清TC、TG、LDL均明显升高(P0.01),HDL明显降低(P0.05),与模型组比较,中西药组血脂无明显变化;5周时,中药组、西药组血清中的TC、TG、LDL开始明显降低,明显低于模型组(P0.05),但仍高于正常组(P0.05)。光镜观察模型组兔股骨头骨组织有骨坏死表现,中西药组病变不明显。结论:脂质代谢是激素诱发股骨头坏死的重要病理基础之一;中药通过调节脂质代谢可以早期预防激素性股骨头坏死;体现其多"靶点"整合调节作用,且毒副作用小。

Study on Prevention of Gushiling Capsule on Hormone-Related Femoral Head Necrosis Based on Curing Before-Disease Theory of Traditional Chinese Medicine

Objective：To investigate the effect of early intervention of Gushuling capsule on hormone-related femoral head necrosis and observe the lipid metabolism change regularity,so as to obtain the new ideas for preventing hormone-induced femoral head necrosis.Methods：Setting up rabbit model of hormone-related femoral head necrosis,experimental rabbits were divided into normal,model,western medicine and Chinese medicine groups,each group was given distilled water or the corresponding medicine,determining the level of serum lipid in each group rabbits after treated 3and 5 weeks,and observing the pathological changes of local femoral head by light microscope.Results：On the third weekend,the serum TC,TG and LDL levels were obviously raised in model,western medicine and Chinese medicine groups,HDL was lowered,the differences were significant to compare with normal group,there were no obvious differences between model and each medicine group,on the fifth weekend,the serum TC,TG and LDL levels were significantly reduced,those were lower than those of model group,but there were differences between the two medicine groups and normal group.Bone tissue of femoral head showed obvious necrosis changes in model rabbits by light microscope,the changes of western and Chinese medicine groups were no obvious.Conclusion：Abnormal metabolism of lipid is an important pathological basis of femoral head necrosis; Traditional Chinese medicine can early prevent femoral head necrosis by regulating lipid metabolism.Traditional Chinese medicine compound predominate over the less side-effect and better integration attribution to the multiple target.
