益智健脑颗粒对SAMP8快速老化小鼠海马BDNF、MMP-9的影响及作用机制研究

摘    要:目的:研究益智健脑颗粒对SAMP8快速老化小鼠海马BDNF、MMP-9的影响及作用机制。方法:6月龄SAMP8快速老化小鼠随机分为中药治疗组和模型组,6月龄SAMR1正常老化小鼠为正常对照组。对各组进行给药治疗,中药治疗组以益智健脑颗粒浓缩液灌胃;正常对照组和模型组以等剂量双蒸水灌胃,共4周。采用双抗体夹心酶联免疫方法检测各纽小鼠海马组织中BDNF、MMP-9的表达。结果:与正常对照组比较,模型组海马BDNF含量明显减少,MMP-9含量明显增多,差异有统计学意义(P〈0.01);与模型组比较,中药治疗组中海马中BDNF含量明显增加,MMP-9含量明显减少,差异有统计学意义(P〈0.01)。结论:益智健脑颗粒可促进小鼠海马神经元合成BDNF,抑制MMP-9产生,这可能是其防治阿尔茨海默病的作用机制之一。

Abstract:Objective:To investigate the effects of Yizhi Jiannao granules (for nourishing the brain) on hippocampal brain -derived neurotrophie factor (BDNF) and matrix metallopeptidase -9 (MMP- 9 ) levels and their action mechanism in senescence -accelerated mouse prone 8 (SAMP8) mice. Methods:SAMP8 mice aged 6 months were randomly divided into traditional Chinese medicine (TCM) treatment group and model group; senescence - resistant 1 ( SAMR1 ) mice aged 6 months were used as normal control group. The TCM treatment group received concentrated solution of Yizhi Jiannao granules by gavage, and the normal control group and model group received an equal volume of double - distilled water by garage. The course of treatment was 4 weeks in both groups. Hippocampal expression of BDNF and MMP - 9 was measured by double - antibody sandwich enzyme - linked immunosorbent assay. Results:Compared with the normal control group, the model group had a significantly decreased hippocampal BDNF level and a significantly increased hippocampal MMP -9 level (P 〈 0. 01 ). Compared with the model group, the TCM treatment group had a significantly increased hippocampal BDNF level and a significantly decreased hippocampal MMP- 9 level (P 〈 0.01 ). Conclusion: Yizhi Jiannao granules can promote synthesis of BDNF and inhibit production of MMP- 9 in the hippocampal neurons of mice ,which may be one of the action mechanisms of Yizhi Jiannao granules in the prevention and treatment of Alzheimer' s disease.
