摘 要: | 目的探讨青蒿素对实验性糖尿病大鼠肾脏蛋白激酶C(PKC)由细胞浆到细胞膜移位活化的抑制作用。方法将实验大鼠随机分为对照组(A组)、糖尿病非治疗组(B组)及糖尿病青蒿素治疗组(C组),B组、C组采用腹腔单剂量注射链脲佐菌素(STZ)65mg/kg的方法建立糖尿病大鼠模型,造模后c组采用青蒿素300mg/(kg·d)腹腔注射。于实验进行到第3周、第6周时分别宰杀大鼠6只,测算肌酐清除率(Ccr)、24h尿白蛋白排泄率(UAER)及肾脏系数(肾脏质量/体质量);采用酶联免疫吸附法(ELISA)测定PKC的活性。结果B组大鼠Ccr、UAER、肾脏细胞膜PKC活性及肾脏系数均较A组明显升高(P〈0.05或0.01);C组大鼠上述指标均较B组明显下降(P〈0.05或0.01)。结论青蒿素通过抑制糖尿病大鼠肾脏细胞PKC移位活化而对糖尿病大鼠早期肾脏病变起到保护作用。

Abstract: | Objective It is to approach the inhibitory effect of artemisinin on the spatiotemporal dynamics activation of protein kinase C (PKC) in the kidney of experimental STZ - induced diabetic rats. Methods The experimental rats were divided into control group (group A) , diabetic without treatment group (group B) and diabetic with treatment of artemisinin group (group C). The model of diabetic rats was established by intraperitoneal injection of a single close of 65 mg/kg induced by STZ. After modeling, the animals in group C were treated with artemisinin of 300 mg/( kg·d) by intraperitoneal injection. UAER, Ccr and Kidney coefficient were observed after 3 - and 6 - week of treatment, while the endogenous PKC activity was measured by ELISA. Results Kidney coefficient, Ccr, UAER and PKC activity at the plasma membrane in group B were significantly higher than those in group A (P 〈 0.05 or 0.01 ); meanwhile, these indexes in group C were markedly lower than those in group B ( P 〈 0.05 or 0.01 ). Conclusion Artemisinin has some renoprotective effects in experimental STZ - induced diabetic rats through inhibiting the spatiotemporal dynamics activation of PKC in the kidney. |
