当归补血汤不同制剂对骨髓抑制小鼠造血调控的实验研究

摘    要:目的]通过比较当归补血汤煎剂及颗粒剂对骨髓抑制小鼠造血机能调控的影响来比较它们之间的疗效差异.方法]本实验采用造血祖细胞培养术、外周血像的计数法、酶联免疫吸附法(ELISA)等方法对造血祖细胞集落数、外周血像的变化及造血微环境中红细胞生成素(EPO)、血小板生成素(TPO)、粒细胞集落刺激因子(GM-CSF)的变化进行了检测.结果]煎剂、颗粒剂能平衡骨髓微环境中EPO、TPO、GM-CSF.的表达;促进骨髓造血细胞从G0/G1期进入G2/M期和S期;促进造血祖细胞增殖,从而提高骨髓抑制小鼠外周血像和骨髓像.结论]当归补血汤煎剂、颗粒剂均能改善骨髓抑制小鼠的造血机能,其中配方颗粒疗效较为突出.

Abstract:Objective] In order to provide some experimental evidences for new herb dosage form, the aim of this experement is to compare the effect of Dang-gui Bu-xue decoction (DGBXT) decoction with that of granule formulation for treating myelosuppression in mice. Methods] Hemopoietic progenitor cell culture and peripheral blood count was used to analyze the hemopoietic function of bone marrow. The hemopoietic factors in bone marrow microenvironment were also tested by ELISA. Results] The ways to improve the account of peripheral blood and nucleated bone marrow cells and to yield the hematopoietic stem / progenitor cells in vitro colony were balancing the expression of cytokine (EPO, TPO, GM-CSF) in bone marrow microenvironment. To facilitate hematopoietic stem/progenitor cells entering the cell cycle is also necessary. But the effect of granule formulation seems to be more better. Conclusions] All DGBXT decoction and the granule formulation can improve the hematopoietic function of bone marrow suppression in mouse. The curative effect of DGBXT granule was more outstanding than that of DGBXT decoction.
