茸菖胶囊对幼年大鼠癫痫后NRSF和BDNF蛋白表达的影响

摘    要:目的]研究茸菖胶囊对幼年大鼠癫痫后神经元限制性沉默因子(NRSF)和脑源性神经营养因子(BDNF)蛋白表达的影响。方法]建立戊四唑点燃癫痫模型,分为中药高、中、低剂量组、西药组、模型组和空白组,用蛋白免疫印迹杂交法检测各组NRSF和BDNF蛋白的变化。结果]幼年大鼠致痫后模型组海马NRSF和BDNF蛋白表达升高,中药高剂量组可显著降低大鼠海马齿状回NRSF和BDNF蛋白表达(P0.01),作用优于西药组和其他中药组。结论]中药复方茸菖胶囊抗癫痫及改善癫痫后认知障碍机制可能与调控组蛋白乙酰化修饰及其所介导的神经元基因表达有关。

Abstract:Objective] To explore the effect of Rongchang capsules(RCC) on the protein expression of neuronal restricted silencing factor(NRSF) and brain-derived neurotrophic factor(BDNF) in hippocampus after pentylenetetrazole(PTZ)induced epileptic seizure in young rats. Methods] To establish the PTZ kindling model of epilepsy, the rats were randomly divided into normal group, model group, valproic acid sodium(VPA)group and RCC low, middle, high dosage group. The expression of NRSF protein and BDNF protein were detected by the Western blot hybridization assay. Results] The expression of NRSF protein and BDNF protein in hippocampus of rats in model groups were significantly higher than those in the blank group, and the difference was statistically significant (P<0.05). Each treat group could reduce the expression, but RCC high dose group was better than VPA and RCC low, middle dosage group(P<0.05). Conclusion] The mechanism of RCC antiepilepsy and improvement cognitive impairment after epilepsy may be related to the inhibition of NRSF and BDNF protein expression in juvenile rat hippocampus after epilepsy.
