丹参注射液对兔急性心肌梗死后心肌细胞凋亡的影响

摘    要:目的]探讨丹参注射液对兔急性心肌梗死(AMI)细胞凋亡的影响及凋亡调控蛋白Fas,Bcl-2表达的影响。方法]用开胸结扎冠状动脉左室支的方法建立急性心肌梗死模型,将已建立心肌梗死模型的日本大耳白兔随机分为两组,模型对照组、丹参组。急性心肌梗死术后中药丹参组静脉推注丹参注射液10日。术后5周取材,对梗死及其周围区进行末端脱氧核苷酸转换酶介导的dUTP-生物素平移末端标记技术对兔实验性急性心肌梗死时梗死及其周围区心肌细胞凋亡情况进行研究,同时观察梗死区内Fas和Bcl-2蛋白的表达。结果]模型对照组和中药丹参组的梗死区及梗死周围区心肌细胞凋亡数均高于正常组(P〈0.05),中药丹参组的梗死区及梗死周围区心肌细胞凋亡数低于模型对照组(P〈0.05)。梗死区及其周围区Fas蛋白的表达,中药丹参组和正常组均低于模型对照组(P〈0.05)。梗死及其周围区Bcl-2蛋白的表达,中药丹参组和正常组均高于模型对照组(P〈0.05)。结论]复方丹参注射液可能通过减少梗死及其周围区Fas蛋白的含量、提高梗死及其周围区Bcl-2的含量来减少急性心肌梗死时心肌细胞凋亡数。

Abstract: Objectives ] Explore the effects of salvia injection on cell apoptosis and the expression of apoptosis regulating gene Fas, Bcl - 2 after acute myocardial infarction in rabbits. Methods ] The model of acute myocardial infarction was made by opening the chest and ligation the left anterior descending and circumflex coronary artery ,The model of acute myocardial infarction in japanese white rabbits were divided randomly into two groups : model group, salvia injection group. The salvia injection group were salvia injection iv bolus ten days after operation, and after five weeks drawing materials. Investigating the Infarction zone and peripheralzone of myocardial cells after acute myocardial infarction in rabbits by the method of terminal deoxynucleotidyl transferase mediated dUTP nick end labeling(TUNEL) ,at the same time observing the expression of Fas and Be1-2 in infaction zone. Results] The model group( given 0.9% sodium chloride after acute myocardial infarction) and apoptosis index of Infarction zone and peripheralzone of myocardial cells of salvia injection group were higher than the normal group(P 〈0.05), apoptosis index of salvia injection group were lower than the model group( P 〈 0.05 ) In the expression of protein Fas in Infarction zone and peripheralzone, salvia injection group and the normal group were both lower than model group ( P 〈 0.05 ). In the expression of protein Bcl- 2 in Infarction zone and peripheralzone, salvia injection group and the normal group were both higher than model group( P 〈 0.05 ). Conclusions ] Compound salvia injection may reduce the Apoptofic Index of myocardial cells when acute myocardial infarction by decreasing the contents of protein Fas in Infarction zone and peripheralzone, and increasing the contents of protein Bcl -2 in Infarction zone and peripheralzone.
