加减补肺汤对COPD“肺气虚证”大鼠血浆SOD活性、MDA含量的影响

目的]采用动物实验方法,旨在对加减补肺汤治疗慢性阻塞性肺疾病(COPD)"肺气虚证"的作用机制进行研究。方法]采用烟熏复合木瓜蛋白酶雾化吸入法复制SD大鼠COPD"肺气虚证"模型。给予加减补肺汤高低剂量及桂龙咳喘宁治疗,观察各组大鼠:一般情况、症状体征、支气管及肺组织病理改变,大鼠血浆SOD、MDA含量的变化。结果]加减补肺汤可以改善COPD"肺气虚证"大鼠全身及呼吸系统症状;能显著改善COPD大鼠支气管、肺组织损伤程度,对组织具有明显的保护作用。与模型组相比,药物治疗的各组血浆SOD活力均明显升高(P<0.05),MDA含量均明显降低(P<0.05)。加减补肺汤高、低剂量组均较桂龙组血浆SOD活力偏高(P<0.05),而MDA含量均较之偏低(P<0.05)。结论]加减补肺汤在一定程度上有保护气道黏膜,减轻肺损伤的作用。可以显著提高血浆SOD活力,降低MDA含量,具有清除自由基及抗脂质过氧化作用。

Effect of decoction for reinforcing lung on the activity of SOD and the content MDA in blood plasma of rats model which have chronic obstructive pulmonary disease(COPD) and TCM syndrome such as "Lung deficiency"

Objective]To study effect of Decoction for Reinforcing lung on rat model which have chronic obstructive pulmonary disease(COPD) and TCM syndrome such as "Lung deficiency".Methods]One hundred rats were randomLy divided into the normal group,the model group,the large dosage of Decoction for Reinforcing lung group,the small dosage of Decoction for Reinforcing lung group and the positive control group (treated by Guilong Kechuanning,a proved effective Chinese patten drug).COPD,Lung deficiency were established on model rat (except normal group),then Decoction for Reinforcing lung was given to the rat.Clinic symptoms and pathological changes of bronchus and lung were observed,as well as the activity of SOD and the content of MDA in in blood plasma were detected.Results]1.General complexion :Rats in model group were quiet and thin,their fur lost smooth and their hair lost seriously;rats in normal group were active and fat,their fur were smooth.Complexions of treat-model group were between the two groups.2.Symotoms:Various degree of chronic cough,catarrh,asthma etc.could be seen in formodel groups gradually.Symptoms were alleviated in treat-model group.3.pathological changes of bronchus and lung tissue:Rats in four model groups had various degree of pulmonary emphysema and extravagated blood,pathological changes of model group were more obviously than other three group.4.The activity of SOD in blood plasma enhanced significantly in low and high dose of Decoction for Reinforcing lung groups.The content of MDA in blood plasma reduced significantly in low and high dose of Decoction for Reinforcing lung groups.Conclusion]Decoction for Reinforcing lung can releave syndromes of rats which have COPD and TCM syndrome such as "Lung deficiency"。Decoction for Reinforcing lung can en-hancing the activity of SOD and reducing the level of MDA in blood plasma.Decoction for Reinforcing lung could clear the free radicals and has the effect of anti-lipidperoxidation.
