左归丸及右归丸对老年大鼠学习记忆相关信号分子表达的影响

摘    要:目的:观察左归丸及右归丸对老年大鼠学习记忆相关信号转导蛋白表达的影响。方法:以自然衰老(24月龄)SD雄性大鼠为动物模型,随机分为老年对照组、老年皮质酮(GC)拮抗组、老年左归组和老年右归组,另设青年对照组(5月龄)。采用Western Blot法观察各组大鼠学习记忆相关信号转导蛋白表达变化,同时观察补肾方药对上述指标异常变化的改善作用。结果:与青年对照组相比,老年对照组大鼠海马组织学习记忆相关信号分子——CaM、pCaMKⅡ、pERK1/2、pCREB及pSynapsinI、Arc蛋白表达均明显降低(P0.05);补肾方药左归丸、右归丸能不同程度地改善上述指标的衰老性变化。结论:补肾方药左归丸及右归丸改善老年大鼠空间学习记忆能力的机制之一可能与延缓学习记忆相关信号分子表达的退行性变化有关。

Abstract:Objective:To observe the effects of Zuogui Wan and Yougui Wan on the expression of learning and memory related signal transduction proteins.Methods:The nature old SD rats were used as senile animal model.The rats were randomly divided into five groups:young group(aged 5 month),senile group(aged 24 month),senile+antagonist RU38486(aged 24 month),senile+Zuogui Wan group(aged 24 month),senile+Yuogui Wan group(aged 24 month).The expression of learning and memory related signal transduction proteins were tested by Western blot method.The regulating effects of kidney-tonifying recipe on above indices were also studied.Results:Compared with young group,the expression of the proteins-CaM,pCaMKⅡ,pERK1/2,pCREB,pSynapsin-1 and Arc were obviously decreased(P<0.05).Zuogui Wan and Yougui Wan,however,could improve the abnormal changes of these indices in senile rats to varying degrees.Conclusion:Zuogui Wan and Yougui Wan could improve spatial learning & momory abilities of senile rats through retarding aging changes of the expression of related signal transduction proteins.
