正交实验法优选黄连-厚朴药对的提取工艺

摘    要:目的:优选黄连-厚朴药对的提取工艺。方法:以小檗碱与厚朴总酚的提取率以及出膏率为指标,采用正交实验法,考察溶剂量,提取次数,提取时间,醇浓度对各指标的影响。运用高效液相色谱法,测定小檗碱,厚朴总酚的含量,流动相:乙腈:0.3%磷酸(三乙胺调节pH=4.0)。结果:黄连厚朴的最佳提取工艺为10倍量药材,70%乙醇回流,提取60min,提取1次。结论:该工艺稳定,可靠,省时,省材。为黄连-厚朴配伍的复方制剂的工艺研究提供了实验依据。

Abstract:Objective: To optimize the extraction process of the couplet medicines Coptis-Magnolia.Methods: Amount of solvent,the extraction time and number of extraction,alcohol concentration on effect of the extraction rate of berberine and the total phenol,the paste rate were evaluated by orthogonal.Determinate the content of berberine and the total phenol by using high performance liquid chromatography(HPLC),the mobile phase: acetonitrile: 0.3% phosphate(adjusting pH to 4 with triethylamine).Results: The best extraction process of the compatibility of Coptis-Magnolia is that they are extracted for 60min with 70% ethanol which is 10 times as much as medicine,and extracting one time.Conclusion: The extraction process is stable,reliable,saving time and materials.The study providing the experimental evidence for the extraction process of couplet medicines Coptis-Magnolia
