甘草黄酮对荷瘤(S180)小鼠免疫细胞数量的影响

摘    要:目的:研究甘草黄酮对荷瘤(S180)小鼠免疫细胞数量的影响,探讨甘草黄酮抗肿瘤作用的免疫学机制。方法:取荷瘤(S180)小鼠,随机分4组:生理盐水对照组、甘草黄酮高、中、低剂量组,每组10只,灌胃给药9天后,采用血细胞分析仪计数血液中白细胞、淋巴细胞的数量;流式细胞仪检测T细胞亚群百分率。结果:甘草黄酮最佳抑瘤率为52.3%(高剂量组);甘草黄酮组血液中白细胞总数、淋巴细胞总数均高于生理盐水对照组,高剂量组增加最多,P〈0.01;甘草黄酮给药组CD4+/CD8+T细胞比值明显高于生理盐水对照组,以高剂量组为最高,P〈0.01。结论:甘草黄酮增加荷瘤(S180)小鼠效应免疫细胞的数量,从而达到抗肿瘤作用。

Abstract:Objective: To investigate the anti-tumor effect of Glycyrrhiza flavonoids(GF) and its immunological mechanism.Methods: Mice bearing sarcoma 180(S180) were randomized into 3 GF-treated groups and one control group.The mice in GF-treated groups were perfused with GF.Leukocyte and lymphocyte count were taken by the blood cell analyzer.Flow cytometry was performed to detect the percentages of the T cell subsets.Results: Treatment of GF resulted in the tumor inhibition rates of 52.3%(high dose group).Blood total leukocyte and lymphocyte count in GF treated groups were all higher than that in the control group,and there was the most significant increase of the number of immune cells in the high dose GF group(P<0.01).Compared with the control group,CD4 /CD8 ratio and percentage of lymphocytes in the tumor of the 3 GF-treated groups were all obvous higher,the high dose group show the best effect(P<0.01).Conclusion: GF exerts its anti-tumor effect possibly through promoting the proliferation and differentiation of immune cells and increasing the immune effector cells in the tumorbearing mice.
