补阳还五汤及拆方对脑缺血再灌注SD大鼠模型细胞凋亡及Bcl-2、Bax蛋白表达的影响

摘    要:目的:揭示补阳还五汤及拆方抗脑缺血再灌注损伤的作用机理,并研究方剂配伍的意义。方法:通过四血管结扎法制作SD大鼠脑缺血再灌注模型,观察总方组与拆方黄芪组、活血组药物对模型脑组织细胞凋亡及Bcl-2、Bax蛋白的表达。结果:模型组大鼠脑组织细胞凋亡明显增加,Bcl-2、Bax染色阳性细胞的表达增多,Bax阳性细胞表达更明显。黄芪组、活血组、总方组有抑制模型神经元细胞凋亡的作用,明显增加Bcl-2染色阳性细胞的表达,降低模型大鼠Bax阳性细胞的表达。活血组与黄芪组作用相比,差异无显著性意义(P〉0.05),总方组与黄芪组、活血组作用相比,差异有显著性意义(P〈0.01)。结论:补阳还五汤及拆方具有通过抑制细胞凋亡,达到抗脑缺血再灌注损伤的作用。

Abstract:Objective: To explore the mechanism of Buyang Huanwu Decoction and its separate components in counteracting the cerebral ischemical reperfusion injury,and to evaluate the compatibility of medicine.Methods: SD rat's models of cerebral ischemical reperfusion injury were established by four-cerebral-artery occlusion method,and then the cerebral cells apoptotic and the expression of Bcl-2 and Bax were observed among the Buyang Huanwu Decoction group,the separate components of Radix Astragali group and the blood-activating group.Results: Nerve apoptotic cells in rats of model group rised apparently,and the expression of Bax and Bcl-2 increased,especially of Bax.All of the three groups have the effects of restraining apoptosis,reduced the expression of Bax,and raised the expression of Bcl-2 apparenly.There is no difference between Radix Astragali group and blood-activating group(P>0.05).Compared with Radix Astragali and blood-activating drugs,Buyang Huanwu Decoction have notable effect(P<0.01).Conclusion: Buyang Huanwu Decoction and its separate components can counteract cerebral ischemical reperfusion injury by means of inhibiting apoptosis,regulating and controlling related gene expression.
