电针对脑缺血再灌注损伤小鼠海马神经元凋亡及Bcl-2、Bax表达的影响

摘    要:目的:观察电针对脑缺血再灌注损伤小鼠海马神经元凋亡及相关基因Bcl-2、Bax表达的影响。方法:复制脑缺血再灌注小鼠模型,电针肾俞、膈俞和百会穴,对照组灌胃喜得镇,分别于治疗第1天和第7天后,以流式细胞术(FCM)法检测小鼠海马细胞凋亡率、Bcl-2、Bax表达。结果:术后第1天,模型小鼠海马细胞凋亡率明显增高,术后第7天较术后第1天增高(P〈0.01)。说明海马细胞由于反复脑缺血再灌注而出现凋亡。模型组术后第1天,海马Bcl-2表达明显下降;术后第7天,则明显增高,并高于术后第1天(P〈0.01)。模型组术后第1天和第7天海马Bax表达均增高(P〈0.01)。电针在两个时点均可上调海马Bcl-2表达,下调Bax表达,降低凋亡率(P〈0.01)。结论:电针具有抑制细胞凋亡、保护神经元的作用。

Abstract:Objective:To explore the effects of electropuncture on apoptosis and expressions of Bcl-2 and Bax in hippocampus neurons of cerebral ischemia reperfusion injury mice.Methods: The models of mice were established by repeated cerebral ischemia-reperfusion,and were given electric acupuncture with points of Shenshu,Geshu,and Baihui.The mice of control group were administered intragastrically with hydergine.With method of flow cytometry,the apoptosis rate and expressions of Bcl-2 and Bax in hippocampus neurons of mice on the first and seventh day after treatment were detected.Results: The apoptosis rate of hippocampus cells increased obviously on the first and seventh day after operation(P<0.01),which illustrated that hippocampus cells appeared apoptosis due to repeated cerebral ischemia-reperfusion.The expressions of Bcl-2 in hippocampus neurons decreased on the first day,but increased notable on the seventh day(P<0.01).The expressions of Bax in hippocampus neurons increased notable on the first and the seventh day(P<0.01).Electropuncture could up-regulate the expressions of Bcl-2,lower the expressions of Bax and lower the apoptosis rate of hippocampus cells of mice(P<0.01).Conclusion: The treatment with electropuncture could inhibit apoptosis and protect neurons.
