中药复方提取物HNA-1治疗猴免疫缺陷病毒感染的实验研究

摘    要:目的:研究中药复方提取物HNA-1治疗猴免疫缺陷病毒(SIV)感染的作用。方法:采用SIVmac239毒株静脉感染15只中国恒河猴,待感染10周病毒载量进入平台期后,将动物随机分为高剂量组、低剂量组和模型对照组,分别灌胃给予1.8g.kg-1.d-1和0.6g.kg-1.d-1HNA-1药物以及等容量温开水;给药治疗8周,之后停药观察8周。观察、检测动物的一般情况、血浆病毒载量、CD4+和CD+8T细胞、血细胞学和活检淋巴结病理改变等。结果:模型对照组在观察期间有2例动物死亡,而治疗组动物均存活;且治疗组动物在体质量、CD4+T细胞数上均显著优于模型对照组(P<0.05);活检淋巴结病理检查显示,治疗组动物淋巴组织保存较好,部分原本淋巴组织结构已发生退变甚至耗竭的动物,经治疗后出现了恢复。结论:中药复方提取物HNA-1虽然不能直接起到抗SIV的作用,但可以提高动物体质量、减少动物死亡、提高CD4+T细胞数、改善淋巴结组织结构的病理退变,对SIV感染导致的免疫系统破坏具有一定的保护作用。

Abstract:Objective:To investigate the effect of HNA-1extracted from a Chinese medicine compound,on Chinese rhesus macaques infected by simian immunodeficiency virus(SIV),which was widely used as model of HIV-1 infection.Methods:15 Chinese rhesus macaques were infected with SIVmac239 by intravenous injection.10 weeks post-infection,animals with plasma viral load at set-point were distributed into 3 groups,including high-dose-treatment group(1.8g·kg-1·d-1HNA-1),low-dose-treatment group(0.6g·kg-1·d-1 HNA-1) and control group(same volume of warm water).Corresponding treatments were delivered to monkeys for 8 weeks followed by an 8 weeks period of observation.Results:2 monkeys in control group died as rapid-progressors,while all animals in treatment groups kept alive.Moreover,animals under treatment showed less body weight loss(P<0.05),more CD4+T cells(P<0.05) and ameliorated pathologic changes of lymphoid tissue.Conclusion:Although HNA-1 couldn’t inhibit the replication of SIV,but it could increase animal’s body weight,decrease the mortality rate,elevate the CD4+T cells counts,and ameliorate the degeneration of lymph node.In a degree,these protect animals from defect of immune system caused by SIV infection.
