益肾喘宁汤调节TCR信号通路蛋白Lck、CD4对肾气虚型支气管哮喘的作用机制

摘    要:目的:探讨益肾喘宁汤联合普米克令舒调节T细胞抗原受体(TCR)信号通路蛋白淋巴细胞特异性蛋白酪氨酸激酶(Lck)、表面抗原分化簇4受体(CD4)对肾气虚型支气管哮喘的作用及机制。方法:将40只大鼠随机分为正常组、哮喘组、肾气虚哮喘组、中药组、中西药组。使用ELISA检测大鼠血清睾酮(T)、皮质醇(COR)、甲状腺素(T4)水平。采用PEX100磷酸化抗体芯片检测TCR信号通路蛋白Lck和CD4,分析其与哮喘气道炎症的相关性;Western Blot法检测小鼠肺组织Lck、CD4蛋白表达。结果:与正常组比较,哮喘组和肾气虚哮喘组大鼠肺组织中Lck蛋白表达水平显著上升,CD4蛋白表达水平显著降低(P<0.05)。治疗后,中药组和中西药组中Lck蛋白表达水平降低,CD4蛋白表达水平升高(P<0.05)。其中,中西药组变化最明显。结论:益肾喘宁汤联合普米克令舒治疗肾气虚型支气管哮喘大鼠有明显疗效,其机制可能与TCR信号通路蛋白Lck、CD4的调节相关。

Abstract:Objective: To explore the effect and mechanism of Yishen Chuanning Decoction combined with Pulmicort on regulating T cell receptor(TCR) signaling pathway protein Lck and CD4 on asthma with kidney qi deficiency. Methods: Forty rats were randomly divided into normal group, asthma group, asthma with kidney group qi deficiency, traditional Chinese medicine group and Chinese and western group. Serum levels of testosterone(T), cortisol(COR) and thyroxine(T4) were measured by ELISA. PEX100 phosphorylated antibody chip was used to detect TCR signaling pathway protein Lck and CD4 and analyze their correlation with asthma airway inflammation. Western Blot assay detected Lck and CD4 protein expression in rats lung tissue. Results: Compared with the normal group, the protein expression level of Lck in the lung tissues of asthmatic group and asthmatic with kidney qi deficiency group was significantly increased, while the protein expression level of CD4 was significantly decreased(P<0.05). After treatment, the protein expression level of Lck in the traditional Chinese medicine group and the Chinese and western groups decreased, while the protein expression level of CD4 increased(P<0.05). Among them, the combination of Chinese and western medicine group changed most obviously. Conclusion: Yishen Chuanning Decoction combined with Pulmicort has an obvious effect on the treatment of asthmatic rats with kidney qi deficiency, and its mechanism may be related to the regulation of TCR signaling pathway protein Lck and CD4.
