地黄饮子对转tau基因果蝇AD模型PI3K/AKT/dTOR信号通路基因表达的影响

摘    要:目的观察地黄饮子对转tau基因果蝇AD模型PI3K/AKT/dTOR信号通路基因表达的影响,探讨其防治阿尔茨海默病的药物疗效和作用机制。方法将果蝇分为3组,即空白组、地黄饮子组、模型组。空白组为正常CS果蝇喂普通培养基,地黄饮子组为转tau基因果蝇喂2%浓度地黄饮子培养基,模型组为转tau基因果蝇喂普通培养基,各组均喂养5天。采用果蝇嗅觉记忆测试检测果蝇学习记忆能力,并运用荧光定量PCR法检测地黄饮子对转tau基因果蝇PI3K、AKT、dTOR mRNA表达的影响。结果 1嗅觉记忆测试中,各组进入有纱布障碍的离心管的果蝇数目变化趋势:空白组果蝇数量变化幅度最小,其次是地黄饮子组果蝇,模型组果蝇变化幅度最大。各组进入无纱布障碍的离心管的果蝇数目变化趋势:地黄饮子组果蝇前4 h进入离心管的数量变化速度较慢,4 h后数量变化速度明显高于其他组的果蝇;模型组果蝇进入离心管的数量整个过程比较缓慢,并且低于其他组果蝇;空白组的果蝇开始4 h进入离心管的数量变化较快并高于地黄饮子组果蝇,4 h后数量变化减慢明显并低于地黄饮子组果蝇。2与空白组比较,模型组和地黄饮子组PI3K、AKT、dTOR mRNA表达差异均有统计学意义(P0.05)。地黄饮子组与模型组比较,PI3K、AKT、dTOR mRNA表达增加,差异有统计学意义(P0.05)。结论地黄饮子可以改善转Tau基因果蝇的学习记忆能力,并上调转tau基因果蝇PI3K、AKT、dTOR mRNA的表达。

Abstract:Objective To explore the effects of "Dihuang Decoction" on expression of tua gene of Drosophila AD model of PL3 L/D TOR signaling pathwas genes. Methods Drosophilae were randomized into three groups : blank group, "Dihuang Decoction" group and model group. The blank group was treated by normal CS culture medium; "Dihuang Decoction" group was tan transgenic drosophilae who were fed 2% " Dihuang Decoction" culture medium; model group were fed by normal culture medium, with the course of 5 days. The drosophila olfactory and memory test was used to detect the learning and memory ability of every drosophila. Then the RT-PCR method was used to detect the expression of PI3K mRNA, Akt mRNA, dTOR mRNA of tau transgenic drosophilae. Results (1)Olfactory memory test showed the changes of the number in drosophila: blank group was smallest, followed the " Dihuang Decoction" group, and the model group was biggest. Numbers of drosophilae in centrifuge tube: " Dihuang Decoction" group was slow before the first 4 hours, and after 4 hours, changes in number of drosophilae was quickly than the other groups; drosophilae in model group were slower than that of the other groups; before the first 4 hours, drosophilae in the blank group which entering the centrifuge tube were much more than "Dihuang Decoction" group. After 4 hours, changes in amount of drosophilae in blank group were lower than that of the "Dihuang Decoction" group. (2) Comapred with blank group, there were significant differences in PI3K mRNA, AKT mRNA and dTOR mRNA expression of the model group and "Dihuang Decoction" group (P 〈 0.05 ). Compared with model group, expression of PI3K mRNA, AKT mRNA and dTOR mRNA were enhanced in "Dihuang Decoction" group ( P 〈 0.05 ). Conclusion "Dihuang Decoction" could effectively enhance study' s and memory' s abilities of Drosophila with Tau gene and up-regulate the expressions of PI3K mRNA , AKT mRNA and dTOR mRNA."
