咳敏合剂对咳嗽变异性哮喘小鼠IFN-γ及IL-13含量的影响

摘    要:目的观察中药“咳敏合剂”早期干预治疗对咳嗽变异性哮喘小鼠肺泡灌洗液中白细胞介素-13(IL—13)及干扰素γ(IFN-γ)含量的影响。方珐昆明小鼠随机分为5组,分别为正常对照组、模型组、咳敏合剂高剂量组、咳敏合剂低剂量组、阳性对照组,每组10只。除正常对照组外,其余小鼠用卵蛋白(OVA)联合氢氧化铝凝胶于第7、14天肌注致敏,第15至28天雾吸激发造模。第29天开始对模型组、咳敏合剂低剂量组、咳敏合剂高剂量组、阳性对照组进行药物干预,共干预5天。收集各组小鼠支气管肺泡灌洗液,采用酶联免疫吸附法(ELISA)测定IFN-γ及IL-13的含量,并观察模型小鼠肺组织病理。结果与模型组比较,咳敏合剂高、低剂量组及阳性对照组IFN-γ、IL-13含量差异有统计学意义(P〈0.01)。与阳性对照组比较,咳敏合剂高、低剂量组IL-13含量差异有统计学意义(P〈0.01)。病理检查显示,咳敏合剂高剂量组能有效减轻肺内炎症,与阳性对照组差异无统计学意义(P〉0.05)。结论咳敏合剂可以有效减轻咳嗽变异性哮喘小鼠的气道炎症。

Abstract:Objective To observe the effect of "Kemin Mixture" on the levels of IFN-γ, and IL-13 in mice with cough variant asthma. Methods The mice were randomized into five groups : normal control group, model group, small dose " Kemin Mixture" group, large dose " Kemin Mixture" group and positive control group, 10 in each group. Except for normal control group, the others were sensitized with intramuscular injection of Ovalbumin combined with Aluminum hydroxide respectively on the 7 and 14 day. From 15 to 28 day, the models were established by mist exhaust apparatus. From the 29 day, the model group, small dose " Kemin Mixture" group, large dose "Kemin Mixture" group and positive control group were intervened by medicine for 5 days. The bronchoalveolar lavage fluid was collected in each group, and detected the levels of IFN-γ and IL-13 by ELISA. The lung tissues were observed. Results Compared with model group, there was significant difference in the levels of IFN-γ and IL- 13 in "Kemin Mixture" groups and positive control group ( P 〈 0. 01 ). Compared with positive control group, there was significant difference in the IL-13 level from the "Kemin Mixture" groups (P 〈 0.01 ). The pathological examination showed that large dose "Kemin Mixture" group can alleviate the lung inflammation, and no significant difference was found between large dose "Kemin Mixture" group and positive control group ( P 〉0. 05 ). Conclusion "Kemin Mixture" can alleviate the airway inflammation in mice with cough variant asthma.
