复方鳖甲软肝方对单侧输尿管结扎大鼠肾组织MMP-9/TIMP-1表达的影响

摘    要:目的探讨基质金属蛋白酶9/基质金属蛋白酶组织抑制剂(1MMP-9/TIMP-1)在肾间质纤维化中的表达情况及其复方鳖甲软肝方的调节作用。方法采用单侧输尿管结扎(UUO)动物模型。将大鼠随机分为7组,正常组、假手术组、模型组、苯那普利组和复方鳖甲软肝方低、中、高剂量组。术后4周处死大鼠,观察肾组织病理改变,并应用免疫组织化学方法检测肾组织MMP-9/TIMP-1的表达。结果模型组肾组织表现为纤维化病理改变,正常组及假手术组肾小管上皮细胞、肾间质细胞胞浆有轻度MMP-9/TIMP-1表达;模型组MMP-9表达明显减少而TIMP-1则呈强阳性表达。复方鳖甲软肝方小、中、大剂量组与模型组比MMP-9强阳性表达(P!0.05),TIMP-1的表达较模型组明显减少(P!0.05)。结论复方鳖甲软肝方可通过对肾组织MMP-9/TIMP-1平衡的调节而发挥抗纤维化的作用。

Abstract:To investigate the expression of matrix metalloproteinase-9/tissue inhibitor of matrix metalloproteinase-1(MMP9/TIMP1)in rats with renal interstitial fibrosis and the mechanism of renal regulation of "Fu fang bie jia ruan gan fang".Methods:Unilateral Ureteral Obstruction rats animal model was used.The rats were randomly divided into 7 groups:normal groups,sham operation group,model group,Benazepril treatment group,FFBJRGF low,middle and high dose treatment groups,Renal tissue were examined by light microscopy at the 4th week after operation.Immunohistochemistry was applied to measure the expression of MMP-9/TIMP-1 in the renal interstitium.Results:Fibrosis was observed in the renal interstitium of rats with obstructive nephropathy,The expression of MMP-9/TIMP-1 in the nomal and shame operation group is low in the renal tubular epithellal cell and renal interstitium cell.In the model group the expression of MMP-9 were decreased whereas the expression of TIMP-1 appered markedly positive(P <0.05);In comparison with model group,in the FFBJRGF treatment group the expression of MMP-9 showed markedly positive and the expression of TIMP-1 were depressed greatly(P <0.05).Conclusions:"Fu fang bie jia ruan gan fang"may suppress fibrosis development via regulating MMP-9/TIMP-1 balance.
