冰敷和消肿散外敷对损伤骨骼肌IGF-1 mRNA表达及组织形态的影响

摘    要:目的探讨冰敷和消肿散外敷对损伤骨骼肌IGF-1 mRNA表达及组织形态的影响。方法 86只SD大鼠,取6只作为空白对照组,其余80只随机分为损伤模型组、消肿散治疗组、冰敷组、冰敷消肿散联合组,并采用打击造模的方法造成急性骨骼肌损伤模型。观察损伤1 d、3 d、7 d、14 d损伤组织IGF-1的mRNA表达及组织形态的变化情况。结果各组之间损伤1 d后,无明显差异。损伤3 d、7 d、14 d后,消肿散组和冰敷消肿散联合组IGF-1的mRNA表达均高于损伤模型组(P0.05)。损伤1 d、3 d冰敷治疗组表现与损伤模型组患侧表现基本一致;消肿散和冰敷消肿散联合组组织水肿炎性细胞浸润程度稍轻于损伤模型组患侧。损伤14 d后:损伤模型组患侧腓肠肌局部血肿机化,瘢痕组织形成,横纹肌纤维再生;消肿散组损伤局部血管再生,瘢痕组织与肌管并存;冰敷治疗组肌纤维得到一定程度的修复,但肌小管和再生的横纹肌纤维排列不规则,间或有结缔组织填充;冰敷消肿散联合组水肿与炎症细胞吸收,肌管修复良好,出现排列较整齐致密的肌纤维组织,与正常组织较为接近。损伤后1 d、3 d、7 d、14 d,冰敷消肿散联合组及消肿散组损伤积分低于损伤模型组患侧(P0.05)及冰敷治疗组(P0.05)。结论消肿散能增加损伤组织IGF-1的mRNA表达,促进骨骼肌的修复。而冰敷并不能增加损伤组织IGF-1的mRNA表达。冰敷对于骨骼肌损伤的修复改善不明显,消肿散组损伤局部血管再生,能改善肌肉组织形态修复程度,冰敷消肿散联合组的肌肉修复与正常组织较为接近。

Abstract:  Objective:To investigate the effects of ice compress and external application of Xiaozhong Powder on IGF-1 mRNA expression and histomorphology in injured skeletal muscle. Methods6 rats from 86 SD rats were taken as the blank control group, the other rats were randomly divided into the injury model group, Xiaozhong Powder treatment group, ice compress group, combination group (ice compress and Xiaozhong Powder). The model of acute skeletal muscle injury was established in these 80 rats by hit method. The changes of IGF-1 mRNA expression and histomorphology in injured skeletal muscle were observed after treatment for 1,3, 7 and 14 days, respectively.  Results:There was no statistically significant difference among these groups after treatment for 1 day. After treatment for 3,7 and 14 days, the IGF-1 mRNA expression in the Xiaozhong Powder group and combination group was higher than that in the injury model group (P<0.05). After treatment for 1 and 3 days, the symptom of affected side in the ice compress group was almost the same as that in the injury model group, and the degree of tissue edema and inflammatory cell infiltration in the Xiaozhong Powder group and combination group was slighter than that in the injury model group. After treatment for 14 days, the gastrocnemius on the affected side showed local hematoma organization, scar tissue formation and striated muscle fiber regeneration in the injury model group. The injury in the Xiaozhong Powder group showed local revascularization and coexistence of scar tissue and myotube. The muscle fiber in the ice compress group was repaired to some extent, while the myotube and regenerated striated muscle fiber were irregularly arranged, and filled with connective tissue occasionally. In the combination group, the edema and inflammatory cell infiltration were absorbed, the myotube was well repaired and the muscle fiber tissue was compact and regularly arranged, which was similar to the normal tissue. After treatment for 1,3, 7 and 14 days, the injury scores in the combination group and Xiaozhong Powder group were lower than those in the injury model group (P<0.05) and ice compress treatment group (P<0.05).  Conclusion:Xiaozhong Powder can increase the IGF-1 mRNA expression in the injury tissue and promote the repair of skeletal muscle, while ice compress can not increase the IGF-1 mRNA expression in the injury tissue. The repair and improvement effects of ice compress on skeletal muscle injury is not obvious. Local revascularization is observed in the Xiaozhong Powder group, and the repair degree of muscle morphology is improved. The muscle repair in the combination group is similar to the normal tissue.
