血府逐瘀胶囊对2型糖尿病大鼠胰岛素抵抗影响的实验研究

目的观察血府逐瘀胶囊对2型糖尿病模型大鼠糖脂代谢及胰岛素抵抗的影响。方法将60只雄性wistar大鼠随机分为空白对照组、模型对照组、血府逐瘀组、吡格列酮组。高脂饲料伴小剂量链脲佐菌素(STZ)建立2型糖尿病大鼠模型,分别给予各组大鼠相应的干预措施。在给药的第2、5、8周眶后取血,检测大鼠空腹血糖(FBG)、血脂、血清胰岛素(FINS)及脂联素水平,计算胰岛素敏感指数(ISI)。结果与空白对照组比较,造模后各组各指标差异均有统计学意义(P<0.05,P<0.01)。与模型组比较,毗格列酮组在干预的第2周时TC、LDL-C、ISI,干预的第5周时TC、TG、LDL-C、FBG、FINS、ISI、血脂联素,干预的第8周时TC、TG、LDL-C、FBG、FINS、ISI、血脂联素差异有统计学意义(P<0.05,P<0.01);血府逐瘀组在干预后的第2周时TC,干预的第5周时TC、TG、血脂联素,干预的第8周时TC、TG、LDL-C、FINS、ISI、血脂联素差异有统计学意义(P<0.05,P<0.01)。结论血府逐瘀胶囊能够改善糖尿病大鼠胰岛素抵抗状态,其作用机制可能是通过调节脂质代谢,从而降低外周游离脂肪酸的含量,增加脂联素的水平。

Experimental research of "Xuefu Zhuyu Capsule" on insulin resistance in type 2 diabetic rats

Objective To observe the effect of "Xuefu Zhuyu Capsule" on glucolipid metabolism and insulin resistance in type 2 diabetic rats.Methods Sixty male rats were randomly divided into 4 groups: blank control group,model control group,Xuefu Zhuyu group and Pioglitazone group.High fat diet with low-dose streptozotocin(STZ) was used to establish type 2 diabetic models.The corresponding intervening measures were used to each group.The blood was extracted at the second,fifth and eighth weeks to detect the levels of FBG,blood fats,fasting insulin and adiponectin,and the insulin sensitivity index was calculated.Results After the 2nd week,all the indexes were changed in Pioglitazone group.After the 5th week,the indexes were changed in Xuefu Zhuyu group.At 8th week,the level of FBG in Pioglitazone group was lower than that of in model group(P<0.01);the level of TC in intervene group was lower than the model control group(P<0.01);and level of LDL in medicine group was lower than model control group(P<0.01,P<0.05);the difference of HDL in each group were insignificant(P>0.05);the level of insulin was decreased and the level of insulin sensitivity index was increased(P<0.01).Conclusion "Xuefu Zhuyu Capsule" can improve the insulin resistance in type 2 diabetic rats by regulating lipid metabolism,reducing free fatty acid content and increasing adiponectin levels.
