红景天抑制乳腺癌血管新生的作用及其机制

摘    要:目的通过体外和体内实验观察红景天对肿瘤血管新生及其相关因子的作用,探索可能机制。方法应用聚合酶链式反应(PCR)方法,检测红景天提取物对乳腺癌MDA-MB-435细胞血管新生相关因子表达的影响;选择裸鼠乳腺癌MDA-MB-435细胞移植瘤模型,以免疫组织化学染色方法研究红景天提取物对裸鼠移植瘤内血管新生相关因子以及微血管密度的作用。结果在24h时红景天提取物对乳腺癌细胞表达的VEGFmRNA无明显影响;而在44h时红景天提取物可显著抑制肿瘤细胞VEGF表达。在红景天提取物治疗24h后,乳腺癌细胞内达菲抗原(DARC)mRNA的表达增高。红景天治疗组裸鼠移植瘤内VEGF表达的免疫组化H评分值较生理盐水对照组低(P<0.05);而移植瘤DARC的表达较后者增高(P<0.05);治疗组裸鼠移植瘤内的微血管密度亦低于对照组(P<0.01)。结论红景天可能通过抑制乳腺癌细胞的促血管新生相关因子的表达、提高抑制血管相关因子的表达等途径达到抑制肿瘤血管新生的作用。

Abstract:Objective To investigate the effects and its mechenism of Rhodiola on angiogenesis in human breast cancer in vivo and in vitro. Methods The expression of angiogenetic factors in breast cancer MDA-MB-435 cells was detected by polymerase chain reaction; the expression of angiogenetic factors and microvessel density in human breast cancer xenografts were measured by immunohistochemistry staining. Results After Rhodiola extract treatment for 24 h, there was no difference of VEGF expression compared with control group, however, the VEGF mRNA levels were down-regulated after 44 h treatment of Rhodiola. DARC mRNA expression was up-regulated significantly in groups after 24 h Rhodiola extract treatment. The proportion and intensity VEGF expression in Rhodiola-treatment Xenografts was reduced, and the H-score was significantly lower than the control group(P < 0.05). DARC expression in Rhodiola group was up-regulated, and H-score was higher than the control group(P < 0.05). There were fewer newly formed vessels in Rhodiola-treated group(P < 0.01). Conclusion Rhodiola may inhibit tumor angiogenesis through both down-regulating angiogenetic factors and up-regulating anti-angiogenetic factors.
