摘    要:目的:探讨参麦注射液对心肌细胞凋亡通路的影响。方法:原代培养心肌细胞,AngⅡ诱导凋亡。MTT法检测线粒体活性,流式细胞仪检测凋亡率及线粒体膜电位,免疫组化染色检测细胞Bcl-2、Bax、Caspase-3蛋白表达。结果:参麦注射液明显降低细胞凋亡率,增高线粒体活性和膜电位,升高Bcl-2蛋白表达,降低Bax、Caspase-3蛋白表达。结论:参麦注射液可抑制心肌细胞凋亡,其机制与线粒体凋亡通路密切相关。

Abstract:Objectives:To investigate the effects of Shenmai injection on apoptotic pathway.Methods: Cultured cardiomyocytes from neonatal rats were stimulated by AngⅡ.Mitochondrial activity was measured by MTT method.Mitochondrial membrane potential,cardiomyocytes apoptosis were detected by flowcytometry.The expressions of Bcl-2,Bax and Caspase-3 protein were detected by immunohistochemical assay.Results: Shenmai injection enhanced mitochondrial activity,mitochondrial membrane potential,significantly increased the level of protein expression o f Bc-l 2,decreased the level of protein expression of Bax and Caspase-3.Conclusions: Shenmai injection can decrease cardiomyocytes apoptosis through interfering with mitochondria-dependent apoptotic pathway.
