益气通络方对缺氧再给氧心肌细胞bcl-2基因表达影响的血清药理学研究

摘    要:目的:观察益气通络方含药血清对缺氧再给氧心肌细胞bcl-2基因表达的作用。方法:采用流式细胞仪检测缺氧再给氧心肌细胞bcl-2基因表达强度。结果:1.缺氧24h、再给氧4h组心肌细胞bcl-2阳性表达比率增加,与正常对照组比较具有显著差异(P<0.01);2.益气通络方低、中、高剂量含药血清组bcl-2阳性表达比率较单纯缺氧再给氧组增加,组间比较呈显著差异(P<0.01)。讨论:缺氧再给氧能诱导心肌细胞bcl-2基因表达,益气通络方含药血清可增强其表达,可能是益气通络方保护缺氧再给氧心肌细胞,发挥抗缺血再灌注损伤的重要机制之一。

Abstract:Objective:To elucidate whether YQTLD can enhance bcl 2 expression of cardiac myocyte during hypoxiareoxygenation. Methods:cultured neonatal rat cardiac myocyte was treated with 24h hypoxia and 4h reoxygenation, the level of bcl 2 expression was detected by fluorescent immunocytochemistry and assayed by flow cytometry. Results:Experiment showed hypoxia reoxygenation could induce bcl 2 expression, which could be further enhanced by serum containing YQTLD. Conclusion:It suggest that YQTL may exert benificial effect on cardiac myocyte during hypoxiareoxygenation, this underlies one of its mechanisms to treat ischemic heart disease.
