白英乙醇提取物诱导人肝癌SMMC-7721细胞凋亡及其对凋亡相关基因表达的影响

摘    要:目的:探讨白英乙醇提取物对人肝癌SMMC-7721细胞凋亡及凋亡相关基因survivin和caspase-3表达的影响。方法:用MTT法检测细胞增殖抑制率,TUNEL法检测细胞凋亡率,半定量RT-PCR检测survivin和caspase-3 mRNA表达。结果:与正常对照组相比,白英提取物各浓度组细胞抑制率显著上升(P<0.001),细胞凋亡率明显升高(P<0.01),Caspase-3 mRNA表达显著上升(P<0.05或P<0.01),Survivin mRNA表达显著下降(P<0.05或P<0.01)。结论:白英乙醇提取物通过上调caspase-3和下调survivin基因表达,诱导细胞凋亡,从而抑制SMMC-7721细胞增殖。

Abstract:Objective: To explore the effects of ethanol extracts of Solanum lyratum Thunb (ST) on human liver cancer SMMC-7721 cells and the expression of apoptosis associated genes survivin and caspase-3.Methods: MTT assay was used to evaluate the proliferation inhibitory rate,and TUNEL method was employed to determine the cell apoptosis rate.Then,the expression of survivin and caspase-3 mRNA was detected by semi-quantitive RT-PCR.Results: compared with that of the control group,the inhibitory rate was increased obviously (P<0.001),and the apoptotic rate was increased markedly (P<0.01).At the same time,the expression of caspase-3 mRNA was increased significantly (P<0.05 or P<0.01) while survivin mRNA was decreased markedly (P<0.05 or P<0.01) in ST extracts groups.Conclusion: ST extracts can induce apoptosis by up-regulating expression of caspase-3 and down-regulating expression of survivin genes,and can inhibit the development of SMMC7721 cells.
