盐杜仲对去卵巢大鼠骨代谢生化指标、骨密度及生物力学的影响

摘    要:目的:观察中药杜仲对去卵巢大鼠骨代谢、骨密度及生物力学的影响,探讨杜仲对骨质疏松症的防治作用。方法:将80只雌性SD大鼠随机分成4组:假手术组、模型组、阿尔法骨化醇组和杜仲实验组。除假手术组打开腹腔,暴露卵巢后即缝合外,其他3组均摘除双侧卵巢。阿尔法骨化醇组和盐杜仲实验组分别用阿法骨化醇软胶囊和盐杜仲灌胃,另外2组正常饲养。3个月后,测定血清雌二醇(E2)、骨钙素(BGP)及尿羟脯氨酸(HOP)。测定股骨骨干重、骨灰分、胫骨抗弯曲力及腰椎骨密度。结果:盐杜仲可增加去卵巢大鼠体内岛含量,降低体内BGP及尿HOP含量,增强去卵巢大鼠腰椎骨密度及提高胫骨抗弯曲力。结论:杜仲可抑制骨吸收,提高骨的强度及质量,对去势大鼠骨质疏松有一定的防治作用。

Abstract:Objective: To observe the effect of Eucommia on bone metabolism, bone density and biomechanics of ovariectomized rats, so as to explore its preventive and curative effects on osteoporosis.Method: A total of 80 female SD rats were divided randomly into 4 groups: sham operation group, model group, alfacalcidol group and eucommia group.Rats in sham operation group were opened abdominal cavity, exposed and sew up the ovaries.Rats in the other groups were excised double ovaries.Gastric perfusion of alfacalcidol soft capsules and salt eucommia were applied to rats in alfacalcidol group and eucommia group respectively.Rats in the other 2 groups were raised normally.Three months later, we measured the following parameters, including serum estradiol (E2), osteocalcin (BGP), urinary hydroxyproline (HOP), weight of femoral backbone, ashes, tibia-bending force and lumbar bone mineral density.Results: Salt eucommia increased E2 content, reduced BGP and urine HOP contents, and enhanced tibia-bending force and lumbar bone mineral density of ovariectomized rats. Conclusion: Salt eucommia can restrain the bone absorption, improve bone intensity and quality.It has certain preventive and curative effects on osteoporosis of ovariectomized rats.
