激素性股骨头坏死患者股骨近端骨髓成骨活性下降的研究

摘    要:目的:观察激素性股骨头坏死患者股骨近端骨髓体外分化为成骨细胞的改变。方法:取激素性股骨头坏死患者股骨近端骨髓,建立人骨髓间充质细胞诱导分化成骨细胞的培养体系,采用观察细胞学形态、MTT法、细胞内ALP含量检测及矿化结节形成,并与正常组进行比较。结果:与正常组相比,MTT测定细胞增殖、矿化结节染色及细胞内ALP含量测定表明激素性股骨头坏死患者股骨近端骨髓体外分化成骨细胞的能力及活性下降(P〈0.01)。结论:激素性股骨头坏死患者股骨近端骨髓间充质干细胞诱导分化成骨细胞的增殖能力和活性下降。

Abstract:Objective:To investigate the alteration of osteogenic activity of mesenchymal stem cells (MSCs) from the proximal femoral marrow of patients with steroid--induced avascular necrosis of femoral head (SANFH) in vitro. Methods:The marrow was taken from the proximal femur of patients with SANFH, and the cell culture system was established to induce MSCs to differentiate into osteoblasts in vitro. The MSCs differentiation activity was measured through cell observation, MTT, intracellular ALP assay and mineralization nodules compared to control cells. Results: MTT, ALP content and calcium nodules studies indicated a significant decrease of osteogenic differentiation of MSCs in the proximal femur in SANFH (P〈0.01). Conclusion: The osteogenic differentiation activity of MSCs into osteoblasts from the proximal femur marrow of SANFH decreased in vitro.
