| 摘 要： | 目的:研究接骨散含药血清对体外培养骨髓间充质干细胞(rBMSCs)增殖与成骨性分化的影响。方法:采用高、中、低剂量灌服大鼠制得接骨散含药血清,以灌服等体积生理盐水制得对照血清;采用贴壁筛选法培养大鼠骨髓间充质干细胞,并以3种不同浓度的含药血清干预第二代rBMSCs。MTT法检测细胞增殖情况;于干预后的第3天、6天、9天和12天分别测定细胞碱性磷酸酶(alkaline phosphatase,ALP)活性、碱性磷酸酶阳性克隆数(CFU-FALP)、钙盐沉积量以及钙化结节数,并比较各组间差异。结果:中剂量接骨散含药血清强烈刺激细胞增殖活性并显著促进骨髓间充质干细胞成骨性分化,表现在该组的ALP活性、CFU-FALP、钙盐沉积量和钙化结节数显著高于对照组(P<0.05)。结论:接骨散含药血清具有促进rBMSCs增殖和成骨性分化活性。
