三七、川芎注射液对脊髓损伤后神经细胞凋亡及脊髓血流量变化的影响

摘    要:目的:本文观察了三七、川芎(sqcx)注射液对脊髓损伤后神经细胞凋亡及脊髓血流量变化的影响。方法:将60只SD大鼠随机分为脊髓损伤后应用三七、川芎注射液组和单纯脊髓损伤组。用改良的Allen's方法造模成功后,分别在术后1,2,4,8,12,24h用Hochesst荧光染色法对脊髓损伤区进行细胞凋亡的检测,采用氢清除法测量脊髓血流量(SCBF)变化。结果:两组中均发现细胞凋亡,模型组损伤8h后损伤节段灰质中阳性细胞达到高峰,12h后灰质中阳性细胞减少,但白质中出现凋亡细胞达到高峰。用药组8h后细胞凋亡数与模型组有明显差异(P〈0.05),24h后两组比较有显著性差异(P〈0.01)。SCBF在损伤后1h明显下降,4h后略有回升,8h后达到最低。24h两组比较有显著性差异(P〈0.01)。结论:脊髓损伤后可导致神经细胞凋亡,三七、川芎注射液能明显抑制细胞凋亡并提高脊髓血流量。

Abstract:Objective: To explore the effect of Sanqi and Chuanxiong Injection (SQCXI) on apoptosis of neurocyte and spinal cord blood flow (SCBF) after spinal cord injury in rats. Methods: Sixty Sprague-Dawley rats were randomly divided into treatment group (treated with SQCXI) and model group. The model of spinal cord injury was established by improved Allen's method. Hochesst fluorescent staining was applied to evaluate apoptosis in neurocyte at 1, 2, 6, 8, 12 and 24 hours respectively after the operation. Meanwhile, SCBF was measured by hydrogen clearance. Results: Apoptosis of neurocyte appeared in both groups. Positively labeled neurocytes in model group reached the peak in the gray matter at 8 hours and decreased at 12 hours meanwhile apoptosis in the white matter reached the peak. There were significant differences in apop- totic neurocytes between treatment group and model group at 8 hours and 24 hours (P〈0.05, P〈0.01, respectively). SCBF decreased at 1 hour, increased moderately at 4 hours, then decreased to the lowest level at 8 hours and apparent sta- tistical difference between the two groups was obtained at 24 hours (P〈0.01), Conclusion: Neurocyte apoptosis existed in spinal cord injury while SQCXI could remarkably inhibit apoptosis and increase the SCBF after spinal cord injury.
