咳喘宁胶囊对慢性支气管炎大鼠血清、肺组织及支气管肺泡灌洗液TNF和IL-1β含量的影响

摘    要:目的:探讨咳喘宁胶囊对实验性慢性支气管炎治疗作用的内在机制。方法:采用改良烟熏法复制大鼠慢性支气管炎模型,实验分组为模型组、咳高组、咳低组、桂龙组,并设不加处理的正常组。以高、低剂量咳喘宁和桂龙咳喘宁进行干预,测定各组大鼠血清、肺组织及支气管肺泡灌洗液(BALF)中肿瘤坏死因子(TNF)和白细胞介素-1(IL-1β)的含量。结果:模型组大鼠血清、肺组织及BALF中TNF和IL-1β含量均较正常组明显升高(P<0.01)。与模型组相比较,治疗各组血清、肺组织及BALF中TNF和IL-1β含量均明显降低(P<0.05或P<0.01);咳高组血清、肺组织中TNF含量均明显低于桂龙组与咳低组(P<0.01),而3个治疗组对BALF中TNF含量的影响差异无统计学意义(P>0.05);咳高组血清、肺组织及BALF中IL-1β含量均低于桂龙组与咳低组(P<0.05或P<0.01)。结论:TNF和IL-1β在慢性支气管炎的病理生理过程中起着重要作用,咳喘宁胶囊通过降低TNF和IL-1β含量,控制炎症反应。

Abstract:ve: To explore the internal mechanism of Kechuanning Capsule (KC) for the treatment of experimental chron ic bronchitis. Methods: The chronic bronchitis model of rats were established by modified smoking method, and allocated to model group, high - dose KC group , low - dose KC group and Guilong group with normal rats as control. The contents of tumor necrosis factor(TNF) and interleukin 1B(IL- 1B) in serum, lung tissue and broncho - alveolar lavage fluid (BALF) were determined. Results: Compared with control group, the contents of TNF and IL - 1B in serum, lung tissues and BALF of model group, were markedly in creased ( P < 0. 01). Compared with model group, the contents of TNF and IL-1B in various treatment groups were markedly de creased ( P < 0. 05 or P < 0. 01). The contents of TNF in serum and lung tissues of high - dose KC group were lower than that of Guilong group and low - dose KC group ( P < 0. 01). But the con tents of TNF in BALF of 3 treatment groups showed no statistical significance ( P > 0. 05) . The contents of IL - 1B in serum, lung tissues and BALF of high - dose KC group were lower than those of Guilong group and low - dose KC group ( P < 0. 05 or P < 0. 01). Conclusion: TNF and IL - 1B play an important role in the patho-physiologic process of chronic bronchitis and KC can control in flammation through the reduction of TNF and IL - 1B content.
