固本化痰通脉方含药血清对大鼠血管平滑肌细胞增殖及细胞周期素D1的影响

摘    要:目的:观察固本化痰通脉方对大鼠血管平滑肌细胞生长增殖、细胞周期及细胞周期素D1表达的影响,以探讨其作用机制。方法:血小板源生长因子BB(PDGF-BB)刺激体外培养的大鼠血管平滑肌细胞使其快速增殖生长,加入不同浓度的固本化痰通脉方含药血清作用一定时间后,通过MTT法测定其对血管平滑肌细胞生长的抑制率,流式细胞技术测定细胞周期及细胞周期素D1的表达。结果:固本化痰通脉方能明显抑制PDGF-BB刺激的血管平滑肌细胞增殖,还以浓度依赖方式减少S期细胞、阻滞细胞于G0/G1期,并下调细胞周期素D1的表达。结论:固本化痰通脉方能抑制血管平滑肌细胞的增殖生长,阻滞细胞周期的进行,因而可能具有抗动脉粥样硬化的作用,其作用机理可能与调控血管平滑肌细胞细胞周期素D1的活性有关。

Abstract:Objective:To investigate the effect of Guben Huatan Tongmai Recipe(GHTR) on the proliferation,cell cycle and eyclin D_1 activity of vascular smooth muscle cells(VSMCs) in rat in order to find out the mechanism of its action.Methods:In this study,cell culture technique was used in vitro.VSMCs stimulated by platelet derived growth factor BB(PDGF-BB) were treated by GHTR in different concentration for a certain period,then the inhibitory ratio of cell proliferation was measured by MTT,cell cycle distribution and cyclin D_1 activity of the cells were observed by Flow-Cytometry respectively.Results:GHTR could obviously inhibit the proliferation of VSMCs stimulated by PDGF-BB,and it could also decrease S phase cells,increase G_0/G_1 phase cells and down-regulate cyclin D_1 activity of VSMCs in concentration-dependent fashion,and the results were significant compared with the normal serum plus PDGF-BB-treated.Conclusion:GHTR could obviously inhibit the proliferation of VSMCs and may prevent atherosclerosis,which mechanism may be realized by regulating cyclin D_1 activity of VSMC.
