摘    要:目的:利用4种发根农杆菌R15834,R1601,R1000,A4诱导药用植物王不留行产生毛状根,建立王不留行的毛状根培养体系。方法:采用共培养法研究不同外植体、菌株、外源激素、侵染时间、预培养时间和共培养时间对王不留行毛状根诱导率的影响。结果:用王不留行叶片作为外植体,外植体预培养60 h,共培养48 h,发根农杆菌R1601侵染10 min,培养基中添加0.1 mg.L-1吲哚丁酸(IBA)转化率最高。结论:王不留行毛状根诱导成功,为其遗传转化提供了依据。

Abstract:Objective: Hairy root of Vaccaria segetalis were induced by the infection of four Agrobacterium rhizogenes strains R15834, R1601, R1000, A4 and an in vitro culture system of the hairy roots was established. Method: Hairy roots were induced by co-culture.Effects of explants, A. rhizogenes, exogenous phytohormones, infecting time, pre-culture time and co-culture time on growth of hairy root were studied. Result: The highest induction frequency was obtained from blade leafs with 60 h pre-culture and 48 h co-culture which were induced by R1601 for 10 mins.The transformation frequency could be raised by culture media with 0.1 mg·L-1 indole-3-batytric acid (IBA). Conclusion: The successful induction of Vaccaria segetalis hairy roots provided a basis for genetic transformation.
