摘    要:目的:优化黄连黄柏总生物碱的提取工艺条件。方法:通过响应面分析法,以黄连黄柏中季铵总碱得率为指标,对液固比、提取时间、提取次数进行优化,并考察乙酸乙酯萃取纯化条件。结果:黄连黄柏总生物碱提取工艺条件为:沸水浴回流提取,液固比为18.125∶1,提取1h,提取4次,季铵总碱得率为66.44mg·g-1生药。乙酸乙酯萃取纯化条件为:碱调提取液pH至12、1倍萃取剂用量、连续萃取6次,中间体中季铵总碱含量达66.130%。结论:为黄连黄柏总生物碱提取工艺的改进提供了参考。

Abstract:Objective:To optimize the extraction condition of total alkaloids from Rhizoma Coptidis and Cortex Phellodendri Chinensis.Methods:The extraction conditions of ratio of liquid to solid,extraction time and times were optimized by response surface method with the total alkaloids as the selective marker.The purification condition of total alkaloids was also studied.Results: The optimum extraction conditions were as follows: extraction repeated 4times,1 hour every time and ratio of liquid to solid was 18.125:1.The yield of total alkaloids under this condition was 66.44 mg·g-1 The total alkaloids were concentrated by acetic ether extraction under pH 12,with the ratio 1:1 of solvent to extract, also the extraction was repeated 6 times,then the content of total alkaloids was 66.130%.Conclusion:The experiment provides a basis for improvement of extraction condition of total alkaloids from Rhizoma Coptidis and Cortex Phellodendri Chinensis.
