桂蚕沙多糖组分的制备与GC-MS分析

摘    要:目的:制备桂蚕沙的多糖并分析其组分,为专属性鉴定特征的建立奠定基础.方法:桂蚕沙多糖制备流程脱脂后水提醇沉→三氯乙酸除蛋白→大孔吸附树脂纯化→活性炭脱色.桂蚕沙多糖组分的确定采用衍生化后GC-MS分析.结果:桂蚕沙多糖提取率为23.5%.桂蚕沙多糖组分:核糖-木糖-鼠李糖-半乳糖-吡喃葡萄糖-阿拉伯糖(1.97∶3.14∶0.58∶0.92∶5.62).结论:气相色谱-质谱联用技术能够有效的分析桂蚕沙中的多糖组分.

Abstract:Objective: To produce the polysaccharide components from Faeces Bombycis Gui and analyze its composition. Method: Polysaccharide component of Faeces Bombycis Gui was extracted by the methods of water extracting-alcohol precipitating after degreasing, and then removed protein with 10% content of trichloroa cetic acid (TCA), purified with AB-8 macroreticular resin and decolored by active carbon. Finally, the polysaccharide component after derivatization was analyzed by gas chromatography-mass. Result: The yield of crude polysaccharide could attain 23.5%. The polysaccharide of Faeces Bombycis Gui was composed of ribose, xylose, rhamnose, galactose, glucopyranose and arabinose. The proportion of it was 1.97 :3.14 :0.58 :0.92 : 5.62. Conclusion: The GC-MS method could analyze the polysaccharide components of Faeces Bombycis Gui effectively.
