槲皮素对大鼠痛风性关节炎抗炎抗氧化活性研究

摘    要:目的:探讨黄酮类化合物槲皮素对尿酸钠致急性痛风性关节炎模型大鼠踝关节肿胀度的影响,对大鼠血清、肝脏和滑膜中炎症因子和抗氧化活性的影响.方法:雄性SD大鼠48只,体重(200±20)g,随机分为6组,每组8只.分别为空白对照组,模型对照组,吲哚美辛(3.0 mg·kg-1)组和槲皮素(100,200,400 mg·kg-)组.每天ig给药1次,连续7d.第5天给药后1h采用大鼠右后肢踝关节腔内注射注入3.0%尿酸钠溶液100μL制备急性痛风性关节炎模型,造模后2,4,8,12,24,48 h用缚线法测取右后肢小腿踝关节同一部位周径,酶联免疫吸附法、比色法等测定大鼠血清、肝脏和滑膜中炎症因子:白细胞介素-1β(interleukin-1β,IL-1β)、肿瘤坏死因子-α(TN F-oα)、前列腺素(prostaglandin E2,PGE2)含量、脂质过氧化终产物:丙二醛(malondialdehyde,MDA)水平和抗氧化酶:超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性.结果:槲皮素能够显著抑制痛风性关节炎大鼠踝关节肿胀度,抑制炎症因子IL-1β,TNF-α,PGE2含量,降低机体脂质过氧化终产物MDA水平,提高机体抗氧化酶SOD,GSH-Px,CAT活性.结论:槲皮素通过抗炎和抗氧化作用发挥治疗痛风性关节炎功效.

Abstract:Objective:To investigate the effects of the flavonoid quercetin on monosodium urate(MSU) crystal-induced inflammation in rats,as well as the swelling of joint,inflammatory factors and antioxidant activity.Method: Forty-eight male SD rats(200±20) g were divided into six groups with 8 rats each group.Group I(control group) was treated with distilled water,Group II served as model group induced by monosodium urate(MSU) crystal and was treated with distilled water(ig),Groups III-V were treated with quercetin(100,200,400 mg·kg-1 body weight,i g),Group VI MSU crystal-induced rats were treated with indomethacin(3.0 mg·kg-1 body weight,ig).quercetin,indomethacin or distilled water was given orally,once daily for seven consecutive days throughout the experiment.On the fifth day,inflammatory reaction was induced by intra-articular injection of 3% MSU crystal suspension 100 μL inside the ankle joint of the right hind limb.The inflammatory reaction was measured by the circumference of the joint with a tie line method at 2,4,8,12,24,48 h,the tissue homogenate of liver,synovial tissue of joint and serum were used for assaying the inflammatory mediators such as interleukin-1β(IL-1β),tumor necrosis factor(TNF-α),prostaglandin E2(PGE2) and antioxidant status including malondialdehyde(MDA),superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px) with enzyme-linked immunosorbent assay and colorimetry.Result: The quercetin had an inhibitory effect on edema in experimental gouty arthritis in a dose-dependant manner,suppressed MSU crystal-induced chemokine production IL-1β,TNF-α,PGE2,decreased levels of the lipid peroxidation end-product MDA,as well as enhanced the activity of antioxidant enzymes SOD,GSH-Px,CAT.Conclusion: The present results clearly indicated that quercetin exerted a strong effect of treatment on acute gouty arthritis by anti-inflammatory and antioxidant effects.
