北沙参多糖的分离、纯化及其体外免疫活性考察

摘    要:目的:分离、纯化北沙参多糖(Glehniae Radix polysaccharides,GRP)并对各纯化组分(GRP-1,GRP-2,GRP-3)进行结构表征及体外免疫活性研究。方法:采用水提醇沉法获得GRP,Sevage法除蛋白,活性炭脱色素,DEAE纤维素DE-52柱色谱法及葡聚糖凝胶G-75(Sephadex G-75)柱色谱法分离与纯化。采用紫外分光光度法(UV)及1-苯基-3-甲基-5-吡唑啉酮(PMP)衍生化-HPLC对北沙参多糖的含量及其单糖组成进行分析,采用傅里叶红外光谱仪(IR)及多角度激光散射仪对GRP-1进行结构表征及相对分子质量测定,并考察北沙参多糖体外对小鼠脾淋巴细胞增殖活性的影响。结果:GRP经柱色谱分离可获得GRP-1,GRP-2,GRP-3共3种多糖组分,UV测得三者的多糖质量分数分别为96.16%,48.64%和89.73%。其中,GRP-1为具有相对均一分子质量的多糖组分,相对分子质量23.01 k Da,由甘露糖-葡萄糖醛酸-鼠李糖-葡萄糖-半乳糖-阿拉伯糖组成,摩尔比为81.86∶0.12∶0.17∶1 259.7∶0.54∶0.33,红外光谱表明其结构中存在α型糖苷键。体外免疫活性结果表明GRP-1组分对体外脾脏淋巴细胞的增殖具有较好的促进作用,此外GRP-2及GRP-3对脾脏淋巴细胞的增殖也具有一定的推动作用。结论:GRP-1是由多种单糖组成的相对均一的多糖组分,并具有较好的体外免疫活性。
