滋阴地黄丸对兔眼脉络膜组织中 VEGF,bFGF和PEDF表达的影响

摘    要:目的:研究滋阴地黄丸对兔眼脉络膜组织中血管内皮生长因子(VEGF),色素上皮衍生因子(bFGF),碱性成纤维细胞生长因子( PEDF)表达的影响.方法:用激光光凝建立兔眼脉络膜新生血管(CNV)动物模型,ig滋阴地黄丸9.75,39 g·kg -1,连续30 d后取眼球进行免疫组织化学染色,测定脉络膜中VEGF,bFGF及PEDF的表达.结果:模型对照组兔眼脉络膜组织中VEGF,bFGF呈强阳性表达及PEDF的弱阳性表达,滋阴地黄丸低、高剂量组中VEGF,bFGF的表达呈阳性或弱阳性,PEDF则呈阳性或强阳性表达.滋阴地黄丸低、高剂量兔眼脉络膜VEGF的表达为(1.20±0.45),(0.40 ±0.55)分,模型组为(2.60±0.55)分,(P<0.01);低、高剂量组bFGF的表达为(1.40±0.55),(0.40±0.55)分,模型组为(2.80±0.45)分,(P<0.01),低、高剂量组PEDF为(1.60±0.89),(2.80±0.45)分,模型组为(0.40±0.55)分(P<0.05,P<0.01).滋阴地黄丸显著地抑制VEGF和bFGF的表达,诱导了PEDF的表达.结论:滋阴地黄丸通过抑制脉络膜中VEGF,bFGF的表达,诱导PEDF的表达而抑制了CNV的形成.

Abstract:Objective: To study the effects of Ziyin Dihaung Wan(ZDW) on the expression of vascular endothelial growth factor (VEGF), fibroblast growth factor(bFGF)and pigment epitheliumderived factor (PEDF) in rabbit choroids tissue. Method: Chorodial Neovascularization (CNV) model was induced by laser, and then administrated with 9.75, 39 g·kg-1 ZDW for 30 days and then rabbit eyeballs were performed immunohistochemical staining in determination of the expression of VEGF, bFGF and PEDF in choroid. Result: Comparing with the strongly positive expression of VEGF, bFGF and weak expression of PEDF in model group rabbit choroidal tissue, the expression of VEGF and bFGF were positive or weakly positive, PEDF expression was positive or strongly positive in low, high dose ZDW treatment groups. Comparing with model control group,ZDW at 9.75 g·kg-1 dose inhibited the expression of VEGF, bFGF significantly (P<0.01) and induced the PEDF significantly (P<0.05). ZDW at 39 g·kg-1 dose inhibited the expression of VEGF, bFGF and induced the expression of PEDF significantly(P<0.01). Conclusion: ZDW inhibited CNV by inhibiting the expression of VEGF and bFGF and inducing the expression of PEDF in rabbit choroids tissue.
