安胃丸对慢性胃炎大鼠血清炎性因子及胃黏膜核因子-κB表达的影响

目的:研究安胃丸对慢性胃炎大鼠血清炎性细胞因子及胃黏膜核因子-κB p65 mRNA(NF-κB p65 mRNA)表达的影响.方法:采用ig60％乙醇,20 mmol·L -去氧胆酸钠ig,0.05％氨水自由饮用,12周复制大鼠慢性胃炎模型,将造模成功大鼠分为模型组、安胃丸低、中、高组.安胃丸低、中、高组分别以3.06,6.12,12.24 g·kg-1·d-1 ig给药,连续给药30 d,末次给药后禁食16h取血,运用ELISA法检测给药后大鼠血清白介素-2(IL-2)、白介素-4(IL-4)、肿瘤坏死因子-α(TNF-α)含量变化,用原位杂交法检测大鼠胃黏膜核因子-κB p65 mRNA的表达情况.结果:安胃丸组胃黏膜上皮细胞排列整齐,腺体无萎缩现象；安胃丸组大鼠血清IL-4和IL-2水平分别高于、低于模型组,TNF-α水平较模型组显著降低,高、中剂量组大鼠胃窦黏膜NF-κB p65 mRNA的表达低于模型组,接近正常对照组.结论:安胃丸具有保护胃黏膜、减轻黏膜炎症反应的作用,其作用可能与抑制NF-κB p65 mRNA表达,减少炎性因子产生及调节机体免疫功能,维持体液免疫和细胞免疫平衡状态有关.

Effect of Anwei Pill on Inflammatory Cytokines of Serum and Expression of NF-kappa B of Gastric Mucosa in Chronic Gastritis Rats

Objective:To exploe effect of Anwei pill on inflammatory cytokines of serum and the expression of nelclear factor-kappa B(NF-κB) p65 mRNA on gastric mucosa in chronic gastritis rats rats.Method:Rats chronic gastritis model was established by combined administration of 60% ethanol,20 mmol·L-1sodium deoxycholate and 0.05% ammonia water.Aanwei pills was given for 30 day.Levels of interleukin(IL-2),(IL-4),tumor necrosis factor-α(TNF-α) in serum were measured by enzyme-linked immunosorbent assay(ELISA).The expression of NF-κB p65 mRNA in gastric mucosa was detected in situ hybridization.Result:The concentration of IL-4 and IL-2 in Anwei pill group was increased and decreased compared with model group,respectively.TNF-α was decreased.Anwei pill could decrease expression of NF-κB p65 mRNA in gastric mucosa.Conclusion:Anwei pill can protect gastric mucosa and inhibit the inflammation reaction.Its mechanism is probably associated with inhibition of the expression of NF-κB p65 mRNA and regulation of immunity.
