连翘化学成分与抗氧化活性研究

摘    要:目的:对中药连翘进行化学成分的分离与鉴定,并对连翘酯苷A的抗氧化活性进行研究.方法:连翘85%的乙醇提取物经D101大L树脂,以水和不同体积分数乙醇洗脱,60%与30%乙醇洗脱部分采用硅胶柱色谱、凝胶色谱和制备液相等手段纯化;利用理化和谱学分析(NMR、ESI-MS)的方法进行结构鉴定.利用DMPD+法和DPPH法测试连翘酯苷A的抗氧化活性.结果:分离得6个化合物,分别鉴定为连翘酯苷A(forsythoside A)(1)、木通苯乙醇B(calceolarioside B)(2)、芦丁(rutin) (3)、连翘苷(forsythin) (4)、异橄榄脂素(isoolivil) (5)、胡萝h苷(daucosterol)(6).结论:连翘酯苷A有很好的抗氧化活性.

Abstract:Objective: To study the chemical constituents of Forsythia suspensa and antioxidant activity of forsythoside A. Method: The compounds were isolated and purified by column chromatography on silica gel Sephadex LH-20 and preparative HPLC, and their structures were identified by physicochemical properties and spectral analysis. DMPD.+ and DPPH experiments were conducted to examine the antioxidant activity of forsythoside A. Result: Six compounds were isolated from the fruits of F. suspensa. The structures of the compounds were determined as forsythoside A(1), calceolarioside B(2), rutin (3) and forsythin (4), isoolivil (5), daucosterol (6), respectively. Conclusion: Forsythoside A showed excellent antioxidant activity.
