蠲痛饮对大鼠子宫内膜异位症微血管密度的影响

摘    要:目的:探讨蠲痛饮对大鼠子宫内膜异位症(endometriosis,EMs)微血管密度(microvessel density,MVD)的影响和抗血管形成作用。方法:采用自体子宫内膜移植法建立EMS大鼠模型,将60只大鼠分为正常组、模型组、蠲痛饮低、中、高剂量组、丹那唑组6组,分别给予蠲痛饮高、中、低剂量(42.9,14.3,4.8 g.kg-1)和丹那唑(0.021 5 g.kg-1)ig,模型组及空白组按照10 mL.kg-1给予生理盐水ig,持续用药28 d,SP免疫组化法检测各组EMS大鼠在位内膜和异位组织灶中MVD的表达。结果:丹那唑组和高、中、低剂量蠲痛饮减小大鼠异位组织体积分别为(32.96±15.38),(15.75±8.36),(38.16±17.32),(42.91±19.54)mm3,与模型组(89.23±26.27)mm3比较差异有统计学意义(P<0.05)。丹那唑组和高、中、低剂量蠲痛饮降低EMS大鼠异位内膜MVD的计数(18.61±5.89,12.24±4.82,14.36±4.13,16.53±4.56),与模型组(26.43±6.75)比较差异有统计学意义(P<0.05)。结论:蠲痛饮对子宫内膜异位症大鼠异位组织中MVD的表达和新生血管形成均有抑制作用,作用呈剂量依赖性。

Abstract:Objective: To observe the effect of Juantongyin (JTY) on microvessel density(MVD) and antiangiogenesis in ectopic endometrium of rats. Method: To observe the effect of high-dose, middle-dose, low-dose JTY and danazol on MVD of endometriotic rats through setting up endometriosis model with rat self-endometria transplantation. Sixty rats were divided into 6 groups randomly:JTY high-dose group (42.9 g·kg-1), middle-dose group (14.3 g·kg-1), low-dose group (4.8 g·kg-1), and danazol group (0.021 5 g·kg-1),model group and control group (10 mL·kg-1) of saline. Result: There was a marked diversity among model group(89.23±26.27) mm3 and other groups(32.96±15.38),(15.75±8.36),(38.16±17.32),(42.91±19.54) mm3 (P<0.05) of the ectopic tissue volume. There was a marked diversity among model group(26.43±6.75), and other groups (18.61±5.89,12.24±4.82,14.36±4.13,16.53±4.56),(P<0.05) of the expression of MVD. Conclusion: JTY has inhibitory effect on the expression of MVD and angiogenesis in ectopic endometrium of rats, in a dose-dependent manner. JTY may be a new drug of antiangiogenesis in ectopic endometrium.
