溃疡平对乙酸性胃溃疡大鼠胃黏膜血流量及血清ET-1,NO含量的影响

摘    要:目的:观察溃疡平对乙酸性胃溃疡大鼠胃黏膜血流量(GMBF)及血清内皮素-1(ET-1)和一氧化氮(NO)含量的影响。方法:采用改良Okabe法制备大鼠慢性乙酸胃溃疡模型,随机分为:正常组、模型组、溃疡平低、中、高剂量组(1.40,2.80,5.60 g.kg-1 i,g)和盐酸雷尼替丁组(2.83×10-2 g.kg-1 i,g)6组。给药14 d后,运用MP150多道生理记录系统中的LDF100C激光多普勒血流测定模块测定溃疡周边部的GMBF;ELISA法检测血清ET-1的含量;硝酸盐还原酶法检测血清NO含量。结果:与模型组相比,溃疡平高剂量组GMBF明显增多(P<0.05),血清ET-1含量明显降低(P<0.01),血清NO含量明显升高(P<0.05);与盐酸雷尼替丁组相比,溃疡平高剂量组GMBF明显增多(P<0.05),血清ET-1含量明显降低(P<0.01),血清NO含量明显升高(P<0.05)。结论:溃疡平刺激NO的合成和释放,反馈性抑制ET-1的释放,从而增加GMBF,这可能是其加速胃溃疡愈合和改善溃疡愈合质量(QOUH)的作用机制之一。
