芍药苷对利血平诱导抑郁模型的影响

摘    要:目的:研究芍药苷对利血平诱导小鼠抑郁模型的行为以及脑组织中单胺递质含量的影响。方法:动物随机分为6组,即对照组、模型组、盐酸氯米帕明40 mg·kg-1组、芍药苷(50,100,200 mg·kg-1)组,均按10 mL·kg-1灌胃给药。每日1次,连续给药7 d,利用利血平诱导小鼠抑郁模型,观察芍药苷的抗抑郁作用。采用HPLC检测小鼠脑组织中单胺递质含量变化。结果:芍药苷(100,200 mg·kg-1)连续灌胃给药7 d可显著提高小鼠抑郁模型体温(P<0.05,P<0.01),芍药苷(200 mg·kg-1)可改善眼睑下垂(P<0.01)及运动不能(P<0.05);并可明显升高脑组织中去甲肾上腺素(NE)(P<0.01和P<0.001)和5-羟色胺(5-HT)(P<0.05,P<0.001)含量。结论:芍药苷具有一定的抗抑郁作用,其机制可能与调节单胺递质含量有关。

Abstract:Objective:To investigate the effects of paeoniflorin on behavior and the brain tissue content of monoamine neurotransmitters in reserpine-induced depression model in mice. Method: The animals were randomly divided into 6 groups, namely the control group, model group, clomipramine(40 mg·kg-1) group and paeoniflorin (50,100,200 mg·kg-1) group. The drugs were given daily for 7 days. Reserpine-induced depression model was established to evaluate the antidepressant effect of paeoniflorin. The content of monoamine neurotransmitters in mice was measured by the HPLC. Result: Paeoniflorin(100, 200 mg·kg-1,ig 7 day) could significantly increase the body temperature in mice model of depression(P<0.05, P<0.01), improved ptosis (P<0.01)and activity inhibition(P<0.05),significantly increased content of Norepinephrine(NE) (P<0.01, P<0.001)and 5-hydroxytryptamine(5-HT) (P<0.05, P<0.001)in mice. Conclusion: Paeoniflorin has remarkable antidepressant effects, its mechanism may be involved in monoaminergic nervous system.
