青秦液对高尿酸血症模型大鼠关节免疫性病理损伤的修复作用

摘    要:目的观察青秦液对高尿酸血症模型大鼠关节免疫性病理损伤的修复作用。方法将实验动物分为正常组、模型组、阳性药组及青秦液大、中、小剂量组,采用灌服腺嘌呤及乙胺丁醇进行高尿酸血症造模。阳性药组按0.09mg/kg体重灌服秋水仙碱;青秦液大剂量组给予5g/(kg·d)的青秦液灌胃,中剂量组给予2.5g/(kg·d)青秦液灌胃;小剂量组给予1.25g/(kg·d)灌胃。1周后,在双侧踝关节背侧将尿酸钠溶液注射到关节腔中。72h后处死动物,抽取关节腔滑液,应用ELISA法检测大鼠关节腔滑液中的炎性细胞因子白细胞介素-8(IL-8)、α-肿瘤坏死因子(TNF—α)的水平,并取踝关节固定于10%福尔马林中,脱钙后常规石蜡包埋,HE染色,光镜观察其用药前后关节周围炎性细胞浸润情况。结果阳性药组、青秦液3个剂量纽大鼠关节腔滑液中炎性细胞因子IL-8、TNF-α与模型组比较差异均无统计学意义(P〉0.05),但从数据上可以看出,青秦液小剂量组大鼠关节腔滑液中IL-8含量低于模型组、阳性药组及青秦液大、中剂量组,而青秦液3个剂量组大鼠关节腔滑液中TNF—α含量低于模型组及阳性药组,其中尤以青秦液中剂量组为低。光镜观察各用药组关节滑膜增生、周围软组织炎症及关节囊渗出均较模型组减轻。结论青秦液对高尿酸血症大鼠急性反应性关节炎病理损伤具有一定的修复作用。

Repairment Effect of Qingqinye on Joint Immune Pathological Injury in Hyperuricemia Model Rats

Abstract:Objective To observe the repairment effect of Qingqinye on joint immune pathological injury in hyperuricemia model rats. Methods Rats were divided into normal group, model group, positive medicine group, Qingqinye high dosage group, Qingqinye middle dosage group and Qingqinye low dosage group. Model was made with adenine and ethambutol by gavage. Colchicine 0.09 mg/(kg·d) were administered to the rats of positive medicine group by gavage. Qingqinye high dosage group were administered Qingqinye 5 g/(kg·d), middle dosage group were 2.5 g/(kg·d) and low dosage group were 1.25 g/(kg·d) by gavage. After 1 week, sodium urate were injected into joint cavity. 72 hours later, these rats were sacrificed. The level of IL-8 and TNF-α in the hydrarthrosis were determined by ELISA method. Ankle joints were fixed by 10% formalin, decalcified, paraffin embedded and HE Dyed. Pathological section with articular cavity and surrounding tissues were made to observe the changes of inflammatory cell infiltration. Results IL-8, TNF-α of articular cavity fluid in positive medicine group and Qingqinye group has no significant difference with model group (P 〉0.05). But according to the data, IL-8 in Qingqinye low dosage group was lower than model group, positive medicine group, Qingqinye high dosage group and Qingqinye middle dosage group. TNF-α in Qingqinye group was lower than model group and positive medicine group, especially in Qingqinye middle dosage group. Pathological section with articular cavity and surrounding tissues were better in positive medicine group and Qingqinye group. Conclusion Qingqinye has a certain effect for repairing joint immune pathological injury in hyperuricemia model rats.
