艾灸及烟条灸预处理对急性胃黏膜损伤大鼠热休克蛋白和炎性细胞因子的影响

摘    要:目的探讨艾灸对急性胃黏膜损伤炎症反应的抑制机制及比较不同灸材对大鼠胃黏膜损伤的保护作用。方法健康SPF级SD大鼠32只,随机分为对照组、模型组、艾灸组、烟条灸组,每组8只。采用无水乙醇灌胃制备急性胃黏膜损伤模型。艾灸及烟条灸穴位为足三里、中脘。按GUTH法计算胃黏膜损伤指数(UI),HE染色光镜下观察胃黏膜组织形态学变化,免疫组织化学法检测胃黏膜细胞中热休克蛋白(HSP)70的表达,放免法测定血清中炎性细胞因子的含量。结果与对照组比较,模型组UI、血清中炎性细胞因子含量明显升高(P〈0.01),胃黏膜组织炎性浸润明显,HSP70阳性表达稍升高。与模型组和烟条灸组相比,艾灸组UI、血清中炎性细胞因子含量明显降低(P〈0.01或P〈0.05),胃黏膜组织形态较为完好,HSP70表达上调(P〈0.01)。结论艾灸及烟条灸对大鼠急性胃黏膜损伤均具有一定的保护作用,其机制可能与诱导机体产生大量HSP70抑制炎性细胞因子的释放有关。

Abstract:Objective To investigate the effect of moxibustion on acute gastric mucosal injury in inflammatory response and compare the inhibition mechanism of different materials moxibustion on gastric mucosal injury in rats. Methods Thirty-two Sprague Dawley rats were randomly divided into control group, model group, moxibustion group and smoke moxibustion group, 8 rats in each group. Acute gastric mucosal injury model was established by anhydrous ethanol gavage. Ulcer index (UI) was caiculated by GUTH method, gastric mucosai histomorphology changes were observed after HE staining under light microscope, the expression of HSP70 in gastric mucosal cells was detected by immunohistochemistry, and the content of inflammatory cytokines in serum was measured by radioimmunoassay. Results Compared with control group, UI and content of inflammatory cytokines in serum of model group were significantly higher (P〈0.01), while the expression of HSP70 increased slightly, and there was obvious inflammatory infiltration in gastric mucosa. Compared with model group and smoke moxibustion group, UI and content of proinflammatory cytokines in serum of moxibustion group were significantly lower (P〈0.01, P〈0.05), while the expression of HSP70 was significantly increased (P〈0.01), and the tissue morphology was good. Conclusion Moxibustion has protective effect on acute gastric mucosal injury in rats, the mechanism may be related to the high expression of HSP70 inhibiting the release of inflammatory cytokines. s
