通督调神针法对脑卒中后抑郁模型大鼠血浆单胺类神经递质的影响

摘    要:目的:探讨通督调神针法治疗脑卒中后抑郁的可能机制。方法:选择健康清洁级雄性Wistar大鼠60只,月龄7—8月,体重250~300g,经1周适应性喂养后行OPEN—FIELD行为学评分筛选,保留评分相近48只用于实验,电脑随机数字法产生12只作为正常组,其余采用单侧颈总动脉不全结扎引起局灶性缺血合并慢性应激结合孤养复制缺血性脑卒中后抑郁大鼠模型。造模完成后再次筛选并保留PSD大鼠24只,电脑随机数字法分为2组:针刺组和模型组,每组12只。正常组常规饲养,不予任何治疗。针刺组予以针刺百会、水沟、神庭、大椎穴,每日针刺一次,每次40min,20min行针一次,每周6次,连续治疗4周。模型组不予任何治疗。3组大鼠分别于造模完成时、治疗4周时运用ZeaLanga神经行为学评分及蔗糖水消耗实验、敞箱实验进行行为学测定,同时采用生物素双抗体夹心酶联免疫吸附法(ELISA)测定大鼠血浆中单胺类神经递质(5-HT、NE)的含量。结果:造模完成时,模型组和针刺组蔗糖水饮用量、水平及垂直运动得分、血浆中单胺类神经递质的含量均较正常组降低,ZeaLanga神经行为学评分提高,差异均具有统计学意义(P〈0.01,P〈0.05);治疗4周时,与正常组和针刺组比较,模型组大鼠蔗糖水饮用量、水平及垂直运动得分、血浆中单胺类神经递质的含量均降低,ZeaLanga神经行为学评分提高,差异均具有统计学意义(P〈0.05,P〈0.01),而正常组与针刺组比较无统计学差异(P〉0.05)。结论:通督调神针法可以改善PSD模型大鼠的行为,提高其血浆单胺类神经递质含量,提示通督调神针法治疗PSD机制可能是通过调节血浆单胺类神经递质含量。

Effect of Dredging Governor Vessel for Regulating Mentality Acupuncture on Monoamine Neurotransmitters in Rats with Post- stroke Depression

Abstract:Objective:To investigate possible mechanism of PSD treated by acupuncture therapy of dredging governor vessel for regulating mentality. Methods:60 male Wistar rats of clean grade, july to august months of age, the body weight of 300 g ± 50 g, doing OPEN - FIELD behavior score after feeding of one week, the scores of retained 48 Wistar rats had similar produced 12 as normal group by computer randomly, the rest were Ligated with unilateral carotid artery combined focal ischemia associated with chronic stress dependent replication solita- ry ischemic rat model of PSD. After modeling screened again and keep 24 PSD rats, which were randomly divid- ed into two groups: model group, acupuncture group,with 12 rats in each group. The normal group fed by con- ventional without any treatment. The acupuncture group received acupuncture Baihui, Shuigou, Shenting, Dazhui, retaining the needles for forty minutes , once per day, six times per week, continuous treatment for four weeks. The model group was not given any treatment. Three groups of rats were behavior tested using Zea Langa neurological score, sucrose consumption test, open field test and using biotinylated double - antibody sandwich enzyme - linked immunosorbent assay (ELISA) to measure the content of plasma monoamine neurotransmitters (5 -HT, NE) in rats respectively after modeling and after treatment for four weeks. Results:After modeling, the amount of drinking sugar water, horizontal and vertical movement score, plasma levels of monoamine neuro- transmitters in model group and acupuncture group were lower than those in the normal group, Zea Langa neuro- logical score increased. The differences were statistically significant ( P 〈 0.01, P 〈 0.05 ). After treatment for four weeks,in model group rats drinking sugar water volume, the horizontal and vertical motion score mono- amine neurotransmitters in plasma levels were reduced. Compared with the normal group and acupuncture group, Zea Langa neurological score increased. The differences were statistically significant (P 〈 0.05, P 〈 0. 01 ). While the normal group clusion : Acupuncture therapy of and the acupuncture group showed no significant difference ( P 〉 0.05 ). Con- dredging governor vessel for regulating mentality can improve the PSD magic rat model of behavior, improve the plasma monoamine neurotransmitters,whose mechanism may adjust the plasma levels of monoamine neurotransmitters.
